Determination of Genetic Stability in Cacao Plants (Theobroma Cacao L.) Derived from Somatic Embryogenesis Using Microsatellite Molecular Markers (SSR)

Abstract: The clonal propagation of T. cacao by somatic embryogenesis (SE) is a promising approach to multiply elite genotypes. Assessing clonal fidelity in plants regenerated from somatic embryos is the first step toward ensuring genetic uniformity in the mass production of planting material. This study assessed the genetic stability of cacao plantlets propagated by SE and conventional grafting for genotypes CCN51 and TSH565 using 13 SSR. The leaves of in vitro plantlets (IVL) were collected from 6-month-old plants and… Show more

“…In the analysis system, each part of the ES cacao plant production at the biofactory was detailed. The required input and output information to describe the behavior of the considered procedure was collected with the direction of staff belonging to the biofactory of Universidad de Antioquia and researchers with significant experience in ES cacao production [12][13][14][15][16][17] . Production plans and data bill of materials (BOM) for 100.000 batch plant production were realized according to Gronau, (2014) 10 (Supplemental 1 and 2).…”

“…Somaclones can be detected through morphological assessments of the off-type regenerants, biochemical response of explants, fingerprinting with protein or isozymes-based markers, and cytogenetic assessment [ 56 , 57 ]. In addition, more advanced DNA- or transposon-based molecular markers [ 58 ] and the use of next-generation sequencing (NGS) screening have also been successfully applied to detect somaclonal variations in fruit tree breeding.…”
