Determination of di−/Polyamine Oxidase Activity in Plants by an In-Gel Spermidine Oxidation Assay

Abstract: Diamine and polyamine catabolism controls plant development, resistance to pathogens and stress responses. Diamine and polyamine oxidases control the catabolism of diamines and polyamines, respectively. Two major routes of di-/polyamine catabolism exist: the terminal and the interconverting. The in vitro activity of each route is assayed by the colorimetric or chemiluminescent determination of hydrogen peroxide produced by oxidation of di-/polyamine substrates. However, these assays fail to estimate activity o… Show more

“…Amine oxidases are multifunctional enzymes that catalyze the oxidative deamination of polyamines (Cona et al 2006; Phvul.007G056600.1). Polyamine oxidases (PAOs) can be oxidatively deaminated by amine oxidases, leading to ROS-dependent stress signaling (Moschou 2018). One class of amine oxidases is the coppercontaining amine oxidase (CuAO, Liu et al 2020, Phvul.007G056200.1; Phvul.007G056400.2; Phvul.007G056500.1), and it has a confirmed role in defense response (Angelini et al 1993).…”
