To build a robust model for the quantification of pharmaceutical drugs, different regression algorithms and preprocessing techniques are employed. In this study, Raman spectroscopy is used for the analysis of solid dosage formulations of two drugs, cefixime and sitagliptin. Using the different amounts of excipients and active pure ingredients (API), the different concentrations of cefixime and sitagliptin are made. The principal component regression (PCA), partial least‐squares regression (PLSR), and PLSR based on feature selection by variable importance in projection (VIP) scores are used to regress the model to quantify the API concentration of cefixime and sitagliptin in a mixture of excipients. These models are built on both normalized and unnormalized data of cefixime and sitagliptin to check the performance of the constructed models. By comparing errors of all trained models, it is observed that the root mean square error of calibration (RMSEC) and root mean square error of prediction (RMSEP) were minimum in the case of PLSR model construction on unnormalized data of both cefixime and sitagliptin.

Quantification of ciprofloxacin in proprietary Ciproxin tablets and generic ciprofloxacin tablets was made using handheld Raman spectroscopy. Powder dilutions of ciprofloxacin or crushed Ciproxin tablets with excipient(s) were made. The spectra were taken either from the powder dilutions or from pellets compressed from these dilutions and were stored in the instrument library. Methods were created from these powder dilutions: each representing a separate model. The spectra of the tablets were identified within each method using the inbuilt algorithm of the instrument. The results showed that only the models made with the crushed Ciproxin tablets were semi‐quantitative for the analysis of Ciproxin tablets. On the other hand, none of the models could identify generic ciprofloxacin tablets. Thus, the spectra were exported to MATLAB for off‐line quantification. Whereas, univariate regression models were hardly linear, partial least square regression models gave linear calibrations with correlation coefficients above 0.99 and root mean square error of calibration below 3.75% m/m. For the prediction of tablets, the models varied in their results with the one created with ciprofloxacin and lactose giving the most satisfactory results. However, in total the method did not give the pharmacopoeial accuracy of ±5% and stays a semi‐quantitative approach although a satisfactory identity of the constituents was achieved. Cephalosporins are widely used as veterinary and human antibiotics. However, cephalosporin abuse is harmful to human health and causes allergic reactions or antibiotic resistance. We investigate a method featuring Raman spectroscopy and chemometrics to quantify mixture solutions of three typical cephalosporins, namely, ceftriaxone, cefotaxime sodium, and cefazolin sodium. Partial least-squares regression models are built on spectral data that are preprocessed by various methods. With prediction relative errors within 5% and high correlation coefficients of 0.998, we demonstrate that Raman spectroscopy combined with multivariate analysis is feasible for use in the quantitative determination of cephalosporin solutions.
