Determination of Canine .BETA.2-Microgloblin in Plasma and Urine by Enzyme-linked Immunosorbent Assay.

Abstract: ABSTRACT. An enzyme-linked immunosorbent assay was developed for the determination of canine β 2 -microglobulin (β 2 -m) in plasma and urine. The detectable sensitivity for pure canine β 2 -m was 0.05 µg/l and the analytical range was 0.1 to 50 µg/l. The mean analytical recovery when pure canine β 2 -m was added to normal plasma was 101.9%. The mean analytical recovery in the urine was 102.1%. The intra-day variation coefficient was 3.1% in plasma, 4.3% in serum and 1.9% in urine. No difference was found betwe… Show more

“…Because of its small molecular weight, β2-MG has the ability to be freely filtered through the glomerular wall into the tubule, about 99.9% of β2-MG is reabsorbed by lysosomal degradation of amino acids rather than back into the blood; therefore, the normal urinary β2-MG content is small, and only the renal tubular damage and urinary β2-MG will increase. Therefore, it may be one of the sensitive markers of proximal tubule injury [110] and is now being implemented in clinical uses.…”

“…Beta 2‐microglobulin assays were performed in serum and CSF samples by the Laboratory of Veterinary Internal Medicine at Kitasato University in Japan. A standard sandwich ELISA assay was used, based on a previously described technique 16. Wells in each immunoplate were coated with 200 μL of purified IgG and diluted with the coating buffer (0.05 M NaHCO 3 , pH 9.6) to a final IgG concentration of 15 mg/L.…”

“…Reference intervals for β 2 m concentration have been reported in the plasma and urine of clinically healthy dogs,16 but to the authors' knowledge this marker has not been evaluated in any disease state. Furthermore, the protein has not previously been assayed in canine CSF.…”

“…Beta2-microglobulin is a low molecular weight protein (11.8 kDa) that is present on the surface of all nucleated cells (Nakajima et al 2001). It is unstable in acidic urine (Donaldson et al 1989).…”
