Determination of bisphenol A in rat brain by microdialysis and column switching high‐performance liquid chromatography with fluorescence detection

Abstract: A sensitive column switching HPLC-fluorescence detection for determination of bisphenol A (BPA) in rat brain by coupling with microdialysis was developed. A microdialysis probe was inserted into the hypothalamus of rat brain and an artificial cerebrospinal fluid was used for perfusion. BPA in brain dialysate was subjected to a fluorescent derivatization with 4-(4,5-diphenyl-1H-imidazol-2-yl)benzoyl chloride (DIB-Cl), and the excess reagent was removed by a column-switching technique. Separation was carried out… Show more

“…In the present study, results of UPLC measurements demonstrate that there was a significant increase of BPA levels within the CSF after juvenile BPA exposure (Figure 1). This confirms that BPA could pass across the BBB,17 and thus exert toxic effect on the CNS. To assess the effect of juvenile BPA exposure on rat cognitive behaviors, two classic spatial memory test models were selected, including the MWM and Y‐maze tests, which are depended on the hippocampus to explore spatial environments 18.…”

“…The in vivo recoveries of the dialysis probe (R in vitro ) were calculated from four replicate measurements of in vitro recoveries (R in vitro ), in vitro losses (L in vitro ), and in vivo losses (L in vivo ), according to previous reports (Sun et al, 2002;Wada et al, 2008 …”

“…On the other hand, it is possible that DIB-Cl reagent could react with the phenol group under the EDC/pyridine condition. [15][16][17] We tried to use the DIB-Cl reagent for the derivatization of LAS under the EDC/pyridine condition. In this result, the DIB-Cl derivatization of LAS was completed under the EDC/pyridine condition, and in other polyether ionophores solutions with HCPI (Fig.…”
