The author discusses methods for the fluorometric determination of affinity constants by linear and nonlinear fitting methods. This is outlined in particular for the interaction between cyclodextrins and several anesthetic drugs including benzocaine. Special emphasis is given to the limitations of certain fits, and the impact of such studies on enzyme-substrate interactions are demonstrated. Both the experimental part and methods of analysis are well suited for students in an advanced lab.

Zinc phthalocyanines have demonstrated a wide range of applications as photosensitisers, particularly in organic synthesis, photocatalysis and photodynamic therapy. Although the introduction of ionic groups on the macrocycle periphery increases their water solubility, aggregation phenomena remain as a significant drawback for their application. In this study, amphiphilic sulfonatocalix[4]arene‐based micelles were used to promote the monomerization of a cationic phthalocyanine through host–guest interactions. A comprehensive spectroscopic characterization was conducted, varying the concentration of sulfonatocalix[4]arene, and binding parameters were calculated to determine the optimal conditions for complete monomerization of the photosensitizer in water. Under these conditions, the combination of sulfonatocalix[4]arene and the photosensitizer activated its photophysical activity in aqueous media, showing significant singlet oxygen photoproduction, photocatalytic capability and photostability.

Calixarene (CAs) stellen die dritte Generation supramolekularer Wirtsstrukturen dar und sind eine der am häufigsten untersuchten makrocyclischen Gerüststrukturen. Sie bieten aufgrund ihrer einfachen Modifikation (fast) unbegrenzte Möglichkeiten, um große Molekülbibliotheken als Materialbasis für vielfältige biomedizinische Anwendungen zu etablieren. Darüber hinaus beteiligen sich CAs und ihre Derivate an verschiedenen nichtkovalenten Interaktionen zur einfachen Erkennung von Gaststrukturen, einschließlich bioaktiver Moleküle, und sind auch wichtige Bausteine für die Herstellung supramolekularer Architekturen. Im Hinblick auf ihre molekularen Erkennungs‐ und Selbstorganisationseigenschaften werden CAs in großem Umfang in der Biosensorik, dem Bioimaging und im Wirkstoff‐/Gentransport eingesetzt. Weiterhin weisen einige CA‐Derivate biologische Aktivitäten auf und können daher als neue Therapeutika eingesetzt werden. Wir fassen hier die vielfältigen biomedizinischen Anwendungen von CAs zusammen, einschließlich der In‐vitro‐Diagnostik (Biosensorik), In‐vivo‐Diagnostik (Bioimaging) und Therapie.
