Determination of an Effective Housekeeping Gene for the Quantification of mRNA for Forensic Applications*

Abstract: The potential application of mRNA for the identification of biological fluids using molecular techniques has been a recent development in forensic serology. Constitutively expressed housekeeping genes can assess the amount of mRNA recovered from a sample, establish its suitability for downstream applications, and provide a reference point to corroborate the identity of the fluid. qPCR was utilized to compare the expression levels of housekeeping genes from forensic-like body fluid stains to establish the most … Show more

“…Two of the housekeeping genes tested in this study, B2M and UBC, proved to be quite sensitive and were detected in most body fluid stains (Table S8). Moreno et al tested the expression levels of a selection of housekeeping genes and showed B2M to be expressed most consistent among body fluids [48]. Based on our data, these housekeeping genes do not appear to be particularly good positive controls in the sense that they should reliably show positive results when biological material is present.…”

“…Anderson et al previously reported that rRNA 18S was more stable than mRNA ACTB in the same species, 27) while Moreno et al recently showed that beta 2 microglobulin (B2M) was equal to ACTB as a useful housekeeping gene in forensic samples. 28) For this reason we compared the stability of ACTB in saliva, semen, and blood stains at various temperatures, and found that relative Ct values in all body ‰uid stains were signiˆcantly higher at 4°C in the refrigerator. Moreover, relative Ct values increased more in all body ‰uid stains at room temperature (22 25°C) after 6 months than at 4°C or 35°C.…”

“…Due to its conceptual simplicity, sensitivity, specificity, and speed, qPCR applications can be found in a variety of fields, including medicine and the life sciences [1, 2]. In clinical diagnostics, qPCR is broadly used for the detection and quantification of bacterial and viral loads, gene dosage determination, cancer diagnostics, and applications in forensic medicine [3–7]. …”
