Determination of amphetamine, methamphetamine and desmethyldeprenyl in human plasma by gas chromatography/negative ion chemical ionization mass spectrometry

Abstract: A sensitive and specific assay for the quantitative determination of amphetamine, methamphetamine and desmethyldeprenyl in human plasma specimens is described. Electron capture/negative ion chemical ionization gas chromatography/mass spectrometry is used to determine the extracted plasma concentrations of the three target compounds as their N-heptafluorobutyryl derivatives. Quantitation is performed by stable isotope dilution using d6-amphetamine and d6-methamphetamine as internal standards. Selected ion monit… Show more

“…In the postamphetamine condition, amphetamine plasma levels were measured in three arterial samples obtained at 0, 30, and 60 min relative to the PET scan as described previously (Reimer et al, 1993). These data ensured that differences in plasma amphetamine concentration did not bias the radiotracer comparison.…”

“…Amphetamine was quantified as its N-heptafluorobutyryl derivative via gas chromatography/mass spectroscopy utilizing a capillary column with mass spectrometer with simultaneous ion monitoring in the negative chemical ionization mode and reactant gas methane/ammonia (95 : 5) (Reimer et al, 1993). Trideuterated amphetamine was used as the internal standard.…”

“…The optimal GC conditions were determined by using standard solutions containing 1 µg/mL of each drug in chloroform. Buffer solutions containing 0.2, 0.4 and 1.2 mol/L NH 4 OH + NH 4 Cl at pH 9.5 were also prepared for pH adjustment.…”

“…In addition, these instruments can be used to identify unknown or interfering GC peaks and to alert the operator about the presence of unidentifiable peaks. 2 Methamphetamine in urine 3 and plasma, 4 alphaprodine in plasma, 5 codeine in plasma; 6 cocaine in urine 7 and meconium, whole blood and plasma, 8 and lidocaine in plasma 9 have been determined using this technique. In recent years, additional interest has arisen in the determination of these analytes in hair, which can be used to monitor drug abuse and determine whether a drug abuser has been abstaining from the drug.…”
