Determination of a 15437 bp nucleotide sequence around the inhA gene of Mycobacterium avium and similarity analysis of the products of putative ORFs

Abstract: A 15437 bp region encompassing the inhA locus from the Mycobacterium awium chromosome was cloned and sequenced. From the sequencing data generated and the results of homology searches, the primary structure of this region was determined. This region contains four known genes (acnA, fabG, inhA and hemH) and two genes, inwA and inwB, whose products display homology with p60 invasion protein of Listeria monocytogenes. Six proteins encoded by putative ORFs contained an RGD motif (often involved in binding to macro… Show more

“…Table 2 shows that the quantity of H37Rv M. tuberculosis transcripts decreased as a function of M⌽ activation, in agreement with a recent study (21). The M. tuberculosis-complex-specific genes whose expression was analyzed were MT10Sa, a structural gene (36), 35 kDa (27), ahpC (31), Ag85B and Ag85C (14), ESAT6 (35), rpoB (24) invB (19), sigF (15, 7), umaA2 (11), and eis (37). Of these 11 mycobacterial genes in resting M⌽ cDNA, 9 were expressed by strain H37Rv M. tuberculosis.…”

“…Extremely low paralogies in buch and mlep are due to massive decay of genes in these genomes. secreted protein of 49.8 kDa, of which the C-terminal part is highly homologous (72%) to mycobacterial invasins (20). The molecular weight of the mature Nfa34810 is in good agreement with the 43-kDa protein, which plays a dominant role in the adherence and invasion of pulmonary epithelial cells by N. asteroides GUH-2 (21).…”

“…paratuberculosis in infected sheep (14), PepA is likely to be expressed by M. tuberculosis growing in macrophages and to play an important role in facilitating survival. Rv1566c is homologous to the p60 invasion-associated protein of Listeria monocytogenes (37). In L. monocytogenes p60 is a murein hydrolase (65) which also participates in host cell invasion (31).…”
