The incidence of colorectal cancer (CRC) is closely related to dietary factors. Dietary factors greatly affect the structure and function of intestinal microorganisms. In the process of cholesterol bile acid metabolism, the gut microbes participate in the conversion of primary bile acid to secondary bile acid and play an important role in the occurrence and development of CRC. Erchen decoction (ECD) has been widely used in basic research and clinical treatment as a traditional Chinese medical prescription, particularly for patients who are overweight and exhibit dyslipidemia. It is widely used in clinical practice in patients with phlegm syndrome in early colorectal cancer. However, the effects of this treatment on microorganisms and the underlying metabolic mechanisms are still unclear. In this study, a model of high-fat diet and azoxymethane combined with dextran sodium sulfate (AOM/DSS) was established, and ECD intervention was administered to investigate the incidence rate of CRC under different dietary conditions, as well as the effect of ECD intervention on CRC mice fed a high-fat diet. The results showed that the incidence rate of colorectal neoplasms increased under conditions of a high-fat diet, and ECD could reduce the incidence of CRC by regulating abnormal lipid metabolism and improving the structure and function of the intestinal flora.

A new, simple, and sensitive ultraperformance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for quantification of fruquintinib was established to assess the pharmacokinetics of fruquintinib in the rat. The internal standard working solution was added to the plasma sample for extraction before analysis. The Acquity UPLC BEH C18 chromatography column (2.1 mm ×50 mm, 1.7 μm) was used to separated analytes under gradient elution using acetonitrile and 0.1% formic acid as the mobile phase. Positive multiple reaction monitoring modes were chosen to detect fruquintinib and diazepam (IS). The precursor-to-product ion transitions were 394.2 → 363.2 for fruquintinib and m/z 285 → 154 for IS. The current method was linear over the concentration range of 1.0–1000 ng/mL for fruquintinib with a correlation coefficient of 0.9992 or better. The matrix effect of fruquintinib and IS was acceptable under the current method. The intra- and interday precision (RSD%) and accuracy (RE%) were within 11.9% and ±13.7%, respectively. The recovery, stability, and sensitivity were validated according to the United States Food and Drug Administration (FDA) regulations for bioanalytical method validation. The analytical method had been validated and applied to a pharmacokinetic study of fruquintinib in rat.

The objective of this study was to determine the effects of sanshoamides and capsaiciniods on plasma and liver lipid levels and the mRNA expression levels of key receptors involved in cholesterol metabolism in hyperlipidemic rats. A total of 56 three-week-old female Sprague-Dawley rats were assigned to 7 treatment groups based on initial body weight (n = 8 rats per group). With certain combinations of sanshoamides and capsaicinoids significantly increased food intake, reduced lipid levels in blood and liver, improved histological characteristics of a fatty liver, down regulated mRNA expression levels of cholesterol 7-alpha-hydroxylase (CYP7A1), 3-hydroxyl-3methylglutary CoA (HMG-CoA) and Farnesoid X Receptor (FXR) in liver and apical sodium-dependent bile acid transporter, Ileal Bile Acid Binding Protein and FXR in the ileum in hyperlipidemic rats. These results indicated that dietary supplementation with sanshoamides and capsaicinoids reduced blood lipid levels and improved cholesterol metabolism in hyperlipidemic rats.
