Determination of 5-trans-prostaglandin E2 in prostaglandin E2 via high performance liquid chromatography of their p-nitrophenacyl esters on a silver ion-loaded cation exchange resin

Abstract: A high performance liquid chromatographic assay of trace amounts of 5-/rans-PGE2 in PGE2 has been developed. This assay, capable of assaying as little as 0.20% 5-frans-PGE2 with a precision of ±10%, is based on the chromatographic separation of the p-nitrophenacyl esters of these compounds on a silver ion-loaded cation exchange resin. This chromatographic column resolves the PGE2-p-nitrophenacyl esters from those of PGA2, PGB2j PGF1a, and PGF2a.

“…Silica gel, loaded with cadmium by a hydrothermal treatment, was also used to separate aromatic amines (486). Silver-loaded silica gels (7, 177, 268, 290) and ion-exchange resins (313,314,471) separated cis-trans isomers of various compounds as well as electron-donor aromatic compounds in general: see Table II. A variant on this procedure is to use thallium (I), which binds ligands less strongly than silver (I) but reacts much more rapidly, giving better column efficiencies (427).…”

“…An entirely novel approach was the use of ion exchangers loaded with silver ions (Emken et al, 1964). This principle has been applied in the column chromatographic separation of fatty acids and their methyl esters (Adlof et al, 1980;Adlof and Emken, 1985;Adlof, 1988), prostaglandins (Merritt and Bronson, 1976) and pheromones (Houx et al, 1974), and later also in the HPLC separation of pheromones (Houx and Voerman, 1976), and lipids (Christie, 1987(Christie, , 1988. Recently, solid phase extraction columns with a cation exchanger in the silver form were proposed for the prefractionation of unsaturated triacylglycerols (Christie, 1990).…”
