Determination of 2- and 3-MCPD as well as 2- and 3-MCPD Esters and Glycidyl Esters (GE) in Infant and Adult/Pediatric Nutritional Formula by Gas Chromatography Coupled to Mass Spectrometry Method, First Action 2018.03

Abstract: Background: Monochloropropanediol (MCPD) and its fatty acid esters as well as glycidyl fatty acid esters are substances generated during oil refining or food processing. The free form released during digestion, 3-MCPD and glycidol, have shown adverse effects in animal studies. Objective: So far, the available analytical methods have not been validated in a collaborative study for infant and adult nutritional formulas. This manuscript describes a single-laboratory validation method in view of a future multilabo… Show more

“…Infant formula is a unique matrix since it has quite diverse raw materials with varying lipid content. For powdered formula, its structure is very dependent on the ingredients and processing, which causes the unique matrix structure and challenging to extract its lipid (Dubois et al ., 2019). The efficiency of lipid extraction method depends greatly on the compatibility of the sample and its solvent, both physically and chemically.…”

“…The fourth method still used the step of Arisseto et al ., (2017) method, but the solvents replaced with isohexane and acetone (80:20), referring to the solvent application by Wohrlin et al ., (2015). The last method was the lipid extraction method by Dubois et al ., (2019) performed with ethyl acetate solvent.…”

“…Referencin g from published results of other works [13,15] and preliminary experiments, the procedure for extracting fatty acid esters of MCPDs from infant formula was developed as followed: About 1.0 g of homogenized sample, accurately weighed, was dissolved in 15 mL of distilled water and transferred into a 100 mL separation funnel. Before extracting the aqueous liquid with organic solvent, 0.25 mL of solution A was added into the separation funnel, and the funnel was gently shaken in 10 second to mix.…”

“…And because MCPDs in food is considered as a serious health issue, AOAC International has also provided a Standard Method Performance Requirements (SPMR) for any analytical method to be used for determination of 2MCPD, 3MCPD, esters of 2 and 3MCPD as well as glycidyl esters in nutritional formula [14]. AOAC International has adopted the GCMS method for the determination of 2MCPD, 3MCPD and glycidyl esters (GE) in nutritional formula [15]. This method, however, monitors only one ion for each analyte, which may affect the method specificity in some complex matrices.…”
