Determination and Confirmation of the Antiviral Drug Amantadine and Its Analogues in Chicken Jerky Pet Treats

Abstract: In this study, we investigated two methods for the detection of antiviral compounds in chicken jerky pet treats. Initially, a screening method developed to detect many different chemical contaminants indicated the presence of amantadine, 1, in some pet treats analyzed. A second antiviral-specific method was then developed for amantadine and its analogues, rimantadine, 2, and memantine, 3. Both methods used an acidic water/acetonitrile extraction. The antiviral-specific method also included a dispersive sorbent… Show more

“…Analyses of these drugs in different matrices have been reported in the literature. These include determinations of memantine hydrochloride in bulk drug and in tablets by sequential injection analysis , analyses of amantadine in chicken muscle by GC and measurements of memantine, amantadine and rimantadine in animal tissues by HPLC–MS and UHPLC–MS . HPLC with optical detectors (e.g., UV‐visible or fluorescence detectors) usually require some derivatization with labeling reagents (e.g., o ‐phthaldialdehyde and 2,3‐naphtalenedicarboxaldehyde) to enable the analyte detection.…”

“…After the solution was vortexed for 30 sec and centrifuged at 11 963 g for 10 min, the supernatant was collected for HPLC-MS/MS detection. The detection method was optimized based on the methods described in development of the HPLC-MS/MS method for detecting amantadine in chicken (Arndt et al, 2005;Yan et al, 2013;Wu et al, 2014;Zhang & Du, 2014;Turnipseed et al, 2015;Mu et al, 2016).…”

“…Amantadine (AMD), a common antiviral agent, has been widely used to treat viral infections in the poultry industry. , However, illegal abuse of AMD causes the presence of its residue in animal-derived products. Humans consuming food with this AMD residue will produce drug resistance. , Therefore, AMD has been prohibited as an antiviral drug in poultry farming in many countries including China since 2005 .…”
