The present study was undertaken to detect and characterize enteric viruses (rotavirus, astrovirus, reovirus, and coronavirus) in breeder poults. Five turkey breeder flocks were selected. Faecal samples were collected from all flocks at 1 week of age and then every other week until the poults reached 9 weeks of age. The faecal samples were pooled in groups of five. Of the 193 pools (''samples'') tested by reverse transcriptionpolymerase chain reaction, 47.2%, 30.6%, and 10.4% samples were positive for astrovirus, rotavirus, and reovirus, respectively. No coronavirus was detected in any of the samples. Overall, 118 (61.1%) samples were positive for one or more enteric viruses. Of the 118 samples, 70 (59.3%) were positive for a single virus and 48 (40.7%) for a combination of viruses. Phylogenetic analysis based on the polymerase gene showed that astroviruses clustered into two groups with sequence homology ranging from 85.6 to 100% at the nucleotide level. Based on NSP4 gene sequences, rotaviruses clustered in a group and had 96.3 to 99.9% sequence homology at the nucleotide level. The reoviruses, based on their S4 gene sequences, clustered in a single group with sequence homology of 96.9 to 100%. Differing amino acid sequences of all three viruses may affect the antigenicity and/or pathogenicity of these viruses and may merit further study. The presence of two or three different viruses in combination may affect the dynamics of turkey health and disease.

This study aimed to investigate the occurrence of coronaviruses (CoVs) in captive birds placed inside a zoological park in Brazil. The role of captive birds in the epidemiology of CoVs in the tropics is poorly understood. A total of 25 (n ¼ 25) different species were tested for viral RNA using individual fecal samples collected from healthy birds. Reverse transcription-polymerase chain reaction targeting the 3 0 untranslated region was used to detect CoV RNA, and positive samples were submitted for sequence analysis. The phylogenetic search revealed nine mutations in the black shouldered peafowl (Pavus cristatus) CoV sequence, which clustered separately from samples previously described in England. This is the first report on the detection of the CoV genome in captive birds in Brazil.

Abstract:The objective of this study was to develop a direct immunohistochemical assay to detect TCoV antigens in formalin-fixed paraffin-embedded sections prepared from experimentally infected poults. The sections of ileo, ileo-cecal junction and ceca regions from intestine were prepared and submitted to two different primary antibodies, first the non-biotin labeled polyclonal antibody for the indirect method, and second the biotin-labeled polyclonal antibody, both raised against IBV by immunized specific pathogen free chickens. All sections were submitted to immufluorescent assay (IFA), a conventional method, and the results compared. The direct immunohistochemical technique showed a higher frequency of antigen in tissues, especially from the ileo-cecal junction with no difference between results obtained by the conventional method. Finally, the immunofluorescence and all modalities of molecular approaches have been played an important role to the diagnosis and prevention of TCoV infections, although to be precise on infectious disease diagnosis, it is necessary complementary techniques. Here, was standardized the biotin labeled polyclonal antibody as reliable tool to be used as an alternative detection of Turkey Coronavirus.
