Four indole-3-carbaldehyde semicarbazone derivatives, 2-((5-bromo-1H-indol-3-yl)methylene)hydrazinecarboxamide (1), 2-((5-chloro-1H-indol-3-yl)methylene)hydrazinecarboxamide (2), 2-((5-methoxy-1H-indol-3-yl)methylene)hydrazinecarboxamide (3), and 2-((4-nitro-1H-indol-3-yl)methylene)hydrazinecarboxamide (4) were synthesized and characterized by ESI-MS and spectroscopic (FT-IR, 1H NMR, and 13C NMR) techniques. The two-dimensional NMR (in acetone-d6) spectral data revealed that the molecules 1 and 2 in solution are in the cisE isomeric form. This evidence is supported by DFT calculations at the B3LYP/6-311++G(d,p) level of theory where it was shown that the corresponding most stable conformers of the synthesized compounds have a cisE geometrical configuration, in both the gas and liquid (acetone and DMSO) phases. The in vitro antibacterial activity of compounds 1–4 was determined against Gram-positive (Staphylococcus aureus and Bacillus subtilis) and Gram-negative (Pseudomonas aeruginosa and Escherichia coli) bacteria. Among all the tested semicarbazones, 1 and 2 exhibited similar inhibitory activities against Staphylococcus aureus (MIC = 100 and 150 μg/mL, respectively) and Bacillus subtilis (MIC = 100 and 150 μg/mL, respectively). On the other hand, 3 and 4 were relatively less active against the tested bacterial strains compared with 1, 2, and tetracycline.

Objective: This study assessed the prevalence of MRSA, ESBL and VRE in students from four dental schools in Europe. Methods: The hand, tongue and nostrils of the students who treated patients (study group) and who did not treat patients (control group) were sampled. After incubation in TSB and subculturing in the presence of 4 µg/ml oxacillin, positive cultures were identified for Staphylococcus aureus by Mannitol salt agar and agglutination tests. The presence of MRSA was confirmed by specific PCR on the species and on the SSCmec genes. ESBL and VRE were isolated using specific CHROMagar and confirmed using antibiotic sensitivity tests. Results: Of the 879 students who participated in this study (454 students which treated patients, 425 controls) a total of 50 students (5.7%) tested positive for a multi-drug resistant bacterium (MDRB); 13 (1.5%) students tested positive for MRSA, 26 (3.0%) for ESBL and 12 (1.4%) for VRE. No statistically significant differences were found between the students who treated patients compared to the control group for any of the MDRB and study centres, excluding MRSA carriage in the Italian student population. The use of antibiotics the year before sampling, was positively associated with the presence of an MDRB (OR 2.0; 95% Confidence Interval 1.10–3.68; p = 0.02). Conclusion : The risk for MDRB carriage and sequential transmission of MDRB for dental health care students and their patients were acceptably low.

The present study investigated the antimicrobial and antibiofilm potential of biosurfactants derived from Lactobacillus fermentum Lf1, L. fermentum LbS4 and Lactobacillus plantarum A5 against clinical isolates of methicillin‐resistant Staphylococcus aureus (MRSA). The cell wall‐bound and intracellular biosurfactants were extracted by solvent extraction method. Fourier‐transform infrared spectroscopy‐based characterization of biosurfactants revealed the heterogeneous chemical composition involving proteins, fatty acids and carbohydrate moieties in LbS4 and A5, while only the sugar and lipid fractions in Lf1. Fatty acid profiling using Gas chromatography‐mass spectrometry indicated hexadecanoic acid and stearic acid as the predominant fatty acids in the biosurfactants of all these strains. Biosurfactants demonstrated dose‐dependent antibacterial action against MRSA isolates with the highest inhibition zone diameter (30·0 ± 0·0 to 35·0 ± 0·0 mm) recorded at 400 mg ml−1. Biosurfactants showed an excellent staphylococcal antibiofilm activity by preventing the biofilm formation and disrupting the preformed biofilms. Visual inspection through scanning electron microscopy witnessed the biosurfactants‐induced alteration in the cell membrane integrity and subsequent membrane pore formation on staphylococcal cells. Taken together, our findings emphasize the prospects of biomedical applications of biosurfactants as bactericidal and biofilm controlling agents to confront staphylococcal nosocomial infections.
