Detection of specific Mycoplasma conjunctivae antibodies in the sera of sheep with infectious keratoconjunctivitis

Abstract: -The serological cross reactions between Mycoplasma conjunctivae, the etiological agent of infectious keratoconjunctivitis (IKC), and the antigenetically and phylogenetically closely related Mycoplasma ovipneumoniae, which is often found in sheep, were analysed. Cross reacting antigens were identified using sera from sheep with IKC and from sheep of herds known to be free of IKC, as well as rabbit hyperimmune serum specific to the two Mycoplasma species. Cross reactions were predominantly due to the strongly a… Show more

“…The humoral immune response against the M. conjunctivae infection was assessed in sera with an indirect ELISA based on the Tween 20 membrane proteins extract of the M. conjunctivae strain HRC/581T (64). This ELISA showed good sensitivity and specificity for M. conjunctivae antibodies in sheep and has also been validated for alpine chamois (Rupicapra rupicapra) (10,15).…”

“…An indirect enzyme-linked immunosorbent assay (ELISA) method based on the Tween 20 extracted fraction of M. conjunctivae strain HRC/581 T has recently been developed to detect M. conjunctivae antibodies in sera of domestic sheep (7). For the analysis of the sheep sera, serum is diluted 1/10, applied to the plates and analysed using a commercially-available monoclonal antibody directed against ruminant immunoglobulin G (IgG) conjugate to horseradish peroxidase, according to the instructions provided by the manufacturer.…”

“…The value of each sample is expressed as a percentage of a positive reference standard, taking a negative reference serum as zero value. The cut-off level (37%) was set 3 standard deviations higher than the mean value for negative controls according to the test validation (7). At present, studies are being performed to develop and evaluate an ELISA to be used in Alpine chamois (28).…”

“…Recent studies detected several specific antigens of M. conjunctivae, including proteins of 175, 73, 68, 60 and 33 kDa, which have been exploited to develop specific serological tests to detect M. conjunctivae infection (Degiorgis et al, 2000;Belloy et al, 2001). To study the antigenic proteins in more detail, we screened a bacteriophage l-based expression gene library of the M. conjunctivae type strain HRC/581 T using rabbit serum raised against a whole cell preparation of M. conjunctivae.…”
