Detection of sequences of TT virus, a novel DNA virus, in German patients.

Abstract: Recently a novel DNA virus, designated TT virus (TTV), that possibly accounts for some of the cases of liver disease of unknown aetiology, was identified in Japanese patients. Using specific primer pairs for conserved regions, we detected TTV DNA by PCR in 16/84 (19%) German patients awaiting orthotopic liver transplantation because of decompensated liver cirrhosis (of diverse causes); in 4/25 (16%) patients with non-A-G hepatitis; in 1/7 patients with autoimmune hepatitis; and in one intravenous drug user. “…The first publication presenting TTV sequences reported that the TTV genome can be classified into two different genotypes (20). This observation was later confirmed by other researchers (2,9,15,35) and expanded to identify a third TTV genotype (25). Subsequently, the existence of three TTV genotypes was substantiated by several other groups (3,7,10,13,31).…”

“…Other researchers also noticed a unique particular nature of the af060550 sequence and also experienced difficulties in classifying this sequence into one of the known TTV genotypes (3,9). The phylogenetic relationships of these three sequences with different TTV genotypes observed in the present study are very complex ( Fig.…”

“…The phylogenetic tree was constructed by the neighbor-joining method with the reference sequences (N22, TA 278:G1a), (Tx011:G1b), (Ts003: G2a), and (NA004 G2b) previously reported 25 and sequences originating from various geographical areas: United Kingdom, 24 Japan, 25,49 China (AB018447, AB018475), and Germany. 50 Classification into genotypes (indicated by Arabic numbers) was based on an evolutionary distance Ͼ 0.30 and that into subtypes (lower case letters) on a distance Ͼ 0.15. Bootstrap values obtained from 1,000 replicates are given as percentages of the branching forks.…”

“…To date, more than 16 genotypes separated by more than 30% divergence at the nucleotide level have been described (6,7,14,16,18,22), incorporating three hypervariable regions (13). Understanding the origins of such diversity is a fundamental problem in virology.…”
