Twenty-five strains of ciprofloxacin-resistant lactic acid bacteria (LAB; 14 Lactobacillus spp. and 11 Streptococcus thermophiles spp.) isolated from commercial yogurts in China were analyzed in this study. For each of these strains, amino acid changes associated with quinolone resistance-determining regions (QRDRs) were investigated using PCR-based detection methods. The same methodology was used to identify the presence of plasmid-mediated quinolone resistance (PMQR) genes in LAB. Sequencing analyses and an efflux pump inhibition test (using reserpine) were also performed as part of the analysis. Our results showed that typical mutations corresponding to quinolone resistance were found in the QRDRs of LAB strains. Detected mutations included Ser80Leu in parC, and Ser83Leu and Glu87Asp in gyrA. In addition, a Tyr74Phe substitution in parC, which had not previously been reported to be associated with quinolone resistance, was observed in two Lactobacillus delbrueckii subsp. bulgaricus strains. For each of the tested strains, the presence of the efflux pump inhibitor, reserpine, resulted in a two-to eightfold reduction in the minimum inhibitory concentrations (MICs) of ciprofloxacin. However, PMQR genes were not observed in any of the strains analyzed. Our results suggest that mutations in the QRDRs or efflux pump could be involved in ciprofloxacin resistance, and that a combination of these mechanisms may lead to increased ciprofloxacin resistance in LAB strains.

A total of 174 E. coli isolates collected from healthy poultry, bovine and ovine recovered between December 2009 and June 2013 in different geographic location in Tunisia, were assessed and examinated for resistance to antimicrobial agents. Avian isolates showed the highest rates of antibiotic resistance: tetracycline (74.7 %), trimethoprim/sulfamethoxazole and amoxicillin with the same rate of resistance (57 %). Prevalences of resistance to the same four antimicrobials in bovine isolates were 33.3 %, 65 %, 30 %, 28.3 %, respectively. However, for ovine isolates, low resistance rates were observed, except for tetracycline (40 %) and amoxicillin (22.85 %). Only one ESBL-producing isolate from chicken was detected. In addition, seventy-seven (44.2 %) isolates were resistant to three or more classes of antibiotics and were considered multidrug resistant (MDR). Interestingly, avian E. coli isolates were more resistant than bovine and ovine ones. These results provide novel insights into the epidemiological characteristics of poultry, bovine and ovine E.coli isolates in Tunisia, and suggest the need for the prudent use of antimicrobial agents in husbandry and the urgent need to establish a national antibiotic resistance monitoring program.

Introduction: Escherichia coli is a commensal bacterium in humans, animals, and the environment that is one of the microorganisms commonly resistant to antimicrobials. Cooked meat products, which are popular in China, are easily contaminated by E. coli during processing and storage. Methodology: In this study, a total of 75 E. coli isolates from cooked meat products in Henan province, China, were assayed for the presence of and horizontal transfer of class 1 integrons. Results: Class 1 integrons were detected in 11 (14.7%) of these isolates, and contained four groups of resistance gene cassettes, including dfrA17-aadA5, dfrA1-aadA1, dfrA12-orfF-aadA2, and an uncommon array of aacA4-catB8-aadA1. The transfer frequency of selected integron-positve donors ranged from 10 -6 to 10 -4 transconjugants per recipient cell, and the integron-containing DNA from the donors could be transferred to E. coli J53Az r with the transformation frequency of 10 -7 to 10 -5 . Conclusions: Class 1 integrons could be transferred to recipient E. coli J53 by conjugation and natural transformation. These findings suggest the role of commensal E. coli isolates from cooked meats as an important reservoir for integrons and the possible transfer of antimicrobial resistance genes to humans via the food chain.
