Detection of Plantago asiatica mosaic virus in lily hybrid plants (Lilium spp.) in Costa Rica grown from imported bulbs

Abstract: Plantago asiatica mosaic virus (PlAMV) was detected in Costa Rica infecting lily plants showing foliar chlorotic and necrotic streaking. Virus identity was established by TEM, sequencing and ELISA. Pathogenicity was confirmed on Nicotiana benthamiana by mechanical inoculation, and PlAMV was detected by ELISA and RT-PCR.

“…To control plant viral diseases and produce healthy plants free of viruses, the indexing process in lilies focuses on both axillary and apical meristems of the parent plant, the resulting plant, as well as the plant coming from the seed [30,31]. Viruses can be transmitted through the parent plant or the seed itself to the new plant, making indexing essential.…”

“…This is particularly crucial for viruses that can be vertically transmitted from the parent plant to the seed. The specific indexing practices may vary based on the type of virus, the plant species, and the desired level of assurance for producing healthy plants free of viruses [30][31][32]. Additionally, producing healthy seeds that have been indexed and shown to be free of known viruses is a common method for controlling plant viral diseases.…”

“…This was reported first in the Netherlands (EPPO, 2011) and in Chile in 2013 (Vidal et al, 2016). However, soon after its first known occurrence in commercial lily hybrids, PlAMV was detected in commercial lily stocks in many other countries in Europe and around the world (Anderson et al, 2013; Chen et al, 2013; Hammond et al, 2015; Harju et al, 2018; Kim et al, 2015; Li et al, 2017; Montero‐Astúa et al, 2017; Pájtli et al, 2015; Parrella et al, 2015; Xu et al, 2017). Interestingly, the isolates from commercial lily products (“European‐like” isolates) have extremely closely related sequences, quite distinct from those obtained from edible lilies in Japan (Hammond & Reinsel, 2018; Komatsu et al, 2017; Ozeki et al, 2006).…”
