Detection of ochratoxin A in aptamer assay using total internal reflection ellipsometry

Abstract: The current work is a continuation of our research targeted the development of novel optical sensing technologies for detection of mycotoxins. The method of (TIRE) was developed in the last decade as a combination of spectroscopic ellisometry and SPR and was proved to be a highly sensitive analytical tool in bio-sensing particularly attractive for detection of low molecular weight analytes, such as mycotoxins. The use of aptamers as highly specific artificial molecular receptors to ochratoxin A (OTA) in conjun… Show more

“…A similar approach was employed in the study of aflatoxin B 1 [219]. Recently, TIRE has also been coupled with SPR for human immunodeficincy virus diagnostics [220], mycotoxin recognition [221] as well as micromolar immunoglobulin determination [222]. TIRE has also been explored for applications in bioimaging with the help of AuNPs [223].…”

“…Immobilization of monoclonal Ab against AFB 1 allowed detection of 0.01 ng/ml of the analyte by shift of refractive index. Total internal reflectance ellipsometry has been applied for detection of OTA bonded to specific aptamers immobilized on gold-coated glass slices [57]. A similar technique was exploited here for detection of AFB 1 and AFM 1 with specific aptamers immobilized on the surface of golden films [58].…”

“…Aptamers labelled with redox groups were successfully used for electrochemical detection of OTA [23]. Recently, we used similar anti-OTA aptamers, though without label, in combination with the TIRE method, and achieved the detection of OTA down to 0.01 ng/ml concentrations [24]. In the current work, we used non-labeled aptamers (from Microsynth, Switzerland) for AFT B1 having the following nucleotide sequence: with SH group at 5' (or C5) terminal and label-free terminal 3' (or C3).…”
