Detection of molecular markers linked to Ry genes in potato germplasm for marker-assisted selection for extreme resistance to PVY in AAFC’s potato breeding program

Abstract: Molecular markers reported to be linked to extreme resistance (ER) against Potato virus Y (PVY) were evaluated in potato germplasm. YES3-3A and YES3-3B, markers linked to Ry sto , were detected in 'Barbara' and its three descendants that exhibit ER to PVY; RYSC3, a marker linked to Ry adg , was detected in breeding clones NY121 and NY123. Assessment of RYSC3 as a marker for selection for Ry adg -mediated ER validated its efficacy in identification of selections with ER to PVY.Key words: potato, molecular marke… Show more

“…Our result supported the previous finding [16] that the Ry sto gene in 'White Lady' is located on chromosome XII and linked to STM0003 and YES3-3A, the markers widely used for marker-assisted selection in breeding programmes [44,45]. Because Ry-f sto mapped by Flis et al [14] and Song et al [15] is also linked to STM0003 and YES3-3A, there is a possibility that the Ry sto gene in 'White Lady' and Ry-f sto have the same source.…”

“…Homology searches of the reference potato genome positioned the candidate genes in three clusters located at the distal end of the long arm of chromosome XII (58–59 Mb of the DM reference genome, Figure a), consistent with marker locations linked to Ry sto (GP122 564 , Witek et al ., , and YES3, Nie et al ., ). This region of chromosome XII consists of three neighbouring superscaffolds, DMB034, DMB750 and DMB114, one of which (DMB114) was proposed as a likely location for Ry sto (Van Eck et al ., ).…”
