Detection of Lung Cancer Using Plasma Protein Profiling by Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry

Abstract: There are no satisfactory plasma biomarkers which are available for the early detection and monitoring of lung cancer, one of the most frequent cancers worldwide. The aim of this study is to explore the application of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-ToF MS) to plasma proteomic patterns to distinguish lung cancer patients from healthy individuals. The EDTA plasma samples have been pre-fractionated using magnetic bead kits functionalized with weak cation exchan… Show more

“…Sample fractionation can be achieved through different approaches, which include gel electrophoresis (see Table 2), isolectrofocusing or anionic/cationic exchange fractionation protocols. [23,28] One of the most widely used approaches, which has been employed in 18 of the studies analyzed, is represented by protein separation by 2D-GE, followed by MS identification of differential spots individually excised from the gel. In 2D-GE, proteins are separated orthogonally on two dimensions, based on their isoelectric point and molecular mass.…”

“…С помощью времяпролетной масс-спектрометрии с матрично-активированной лазерной десорбцией/ионизацией (MALDI-TOF-MS) можно получить профили экспрессии белков в биологических образцах человека и обнаружить потенциальные биомаркеры рака. Недавно показана эффективность этой технологии для открытия белковых маркеров в сыворотке/плазме крови человека [7][8][9][10][11].…”
