Detection of Inflammation- and Neoplasia-Associated Alterations in Human Large Intestine Using Plant/Invertebrate Lectins, Galectin-1 and Neoglycoproteins

Abstract: Commonly, plant and invertebrate lectins are accepted glycohistochemical tools for the analysis of normal and altered structures of glycans in histology and pathology. Mammalian lectins and neoglycoproteins are recent additions to this panel for the detection of lectin-reactive carbohydrate epitopes and glycoligand-binding sites. The binding profiles of these three types of probes were comparatively analyzed in normal, inflamed and neoplastic large intestine. In normal colonic mucosa the intracellular distribu… Show more

“…Similar to histochemical studies presented in this issue [Brinck et al, 1998;Danguy et al, 1998;Plendl and Sinowatz, 1998;Zschäbitz, 1998] plant and invertebrate lectins have been employed as tools to characterize the oligosaccharide chains of the ZP proteins. As outlined by Gabius [1998] and in this issue by von der Lieth et al [1998], these sugar receptors frequently select a low-energy conformation for ligand binding.…”

“…These saccharide structures can be detected by reactivity with specific plant lectins (41)(42)(43)(44)(45). As susceptibility to galectin-1 is regulated by expression of these saccharide ligands, it was important to characterize the saccharide structures on Sezary cells.…”

“…So far, classical plant lectin histochemistry has been the standard method to access glycan expression and its modulation. Due to fine-specificity differences between plant and mammalian lectins, even between those harboring identical monosaccharide specificity, as seen for example for galactose [Brinck et al, 1998], it is obvious that monitoring actual capacity for establishing recognitive interactions in situ is advanced by using endogenous lectins as test substances.…”
