Detection of in vitro and in vivo cleavage of high molecular weight kininogen in human plasma by immunoblotting with monoclonal antibodies

Abstract: Purified human high-mol-wt kininogen (HMWK), the cofactor of the contact phase of blood coagulation, migrated as a single band (approximately 110,000 mol wt) in a continuous buffer sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), but appeared as two separated bands (approximately 120,000 and 105,000 mol wt) when analyzed in a discontinuous buffer SDS-PAGE system. After elution from SDS polyacrylamide gels, each of the two bands showed coagulant activity. Six murine monoclonal antibodies (M… Show more

“…Hageman factor is associated with b-A deposits [17], HK is massively cleaved in CSF of AD patients [18], and plasminogen [31], plasminogen activator and inhibitors are produced in brain [32,33]. In vitro, soluble b-A has the capacity to induce cleavage of HK [21], in a F.XII-and kallikrein-dependent manner (Fig. 4a), and enhances the t-PAdependent conversion of plasminogen to plasmin (Fig.…”

“…Contact system activation was assessed by measuring the degree of cleavage of high molecular weight kininogen (HK) [20,21], essentially as described for C factors. The HK bands were visualized on blotting membranes with polyclonal goat anti-light chain HK, biotin-conjugated rabbit anti-goat IgG and an avidin-alkaline phosphatase substrate.…”

“…Kallikrein then cleaves high molecular weight kininogen to form BK. 5,6 Additionally, factor XII is a substrate for kallikrein, thus generating a positive feedback loop to accelerate the formation of BK following the initial proteolytic steps. 7 High plasma levels of BK produced during HAE attacks engage the bradykinin 2 receptor (B2R), leading to a complex series of cellular events that disrupt tight junctions between endothelial cells, resulting in vascular leakage that presents clinically as painful swelling.…”
