Detection of IgM antibodies to measles virus by enzyme-immunoassay

Abstract: A method of solid-phase enzyme-immunoassay (EIA) with horseradish-peroxidase-conjugated anti-mu-globulin was used to determine IgM antibodies to measles virus in human sera. Antigens prepared from measles-infected and noninfected Vero cells passively adsorbed to polystyrene cuvettes were both important for the tests, because sera from many patients convalescing from viral infections and sera from rheumatoid arthritis patients were able to bind to control antigen. Sera from measles patients, patients with other… Show more

“…Nonspecific results in the indirect t~l?e of IgM antibody EIAs, caused by either rheumatoid factor' (12,14,16,23,24,27) or antibodies directed against cellular structures (2,14,22,23) are frequently encountered. Vgith the direct EIA (3,17,18,19,21,26) principle these nonspeeifie bindings should be minimal.…”

“…The results for tile serum specimens arriving at the laboratory in the afternoon could be given the next morning. The indirect measles EIA which has been used in our diagnostic unit for many years is performed in a single day, but the specificity tests of the positive specimens by adsorbing (23,24) rheumatoid factor require a day or two more.…”
