Detection ofChlamydia trachomatisandNeisseria gonorrhoeaewith the cobas CT/NG v2.0 test: performance compared with the BD ProbeTec CT Qxand GC Qxamplified DNA and Aptima AC2 assays

Abstract: The cobas CT/NG v2.0 test performs well using urogenital swabs, urine and cervical samples collected in PreservCyt solution.

“…Initial C. trachomatis screening was performed with either Roche Cobas Amplicor or Roche Cobas 4800 (Roche Diagnostics, Basel, Switzerland), which are both very sensitive commercial assays (lower detection limit 40 bacteria/ml 21 , sensitivity 91.2%-98%, [21][22][23] specificity ≥98.8%.) 22 23 The C. trachomatis load was quantified in positive samples by inhouse quantitative PCR (qPCR) targeting the chromosomal OmpA-gene.…”

“…These are the nucleic acid amplification tests (NAATs) based on a polymerase chain reaction (PCR), namely Roche AMPLICOR and COBAS AMPLICOR 2 : one based on strand displacement amplification (BD ProbeTec ET System; Becton Dickinson, Sparks, MD) 3 and one based on transcription-mediated amplification (TMA): APTIMA Combo 2 (Hologic Inc. San Diego, CA). 4 The Cobas CT/NG v2.0 test (Roche Molecular Systems, Branchburg, NJ) 5 behaves comparably to the others mentioned, as does the Abbott m2000 Real Time CT/NG assay (Abbott Molecular; Des Plaines, IL). 6 The GeneXpert CT/NG assay (Cepheid, Sunnyvale, CA), with similar performance to the aforementioned NAATs produces a result within 1.5 h. All these methods offer excellent sensitivity, usually well above 90%, while maintaining very high specificity.…”

“…Appropriate targets could be missed by the software during the process, requiring manual intervention. 54 False-positive amplification due to hybridisation by primers is [192][193][194] (Continued) also common. 55 Hopefully, as the patent license (EP 1020534 B) for this technology expires in 2019, LAMP would experience more modifications.…”
