Ulcerative colitis and Crohn's disease are inflammatory bowel diseases thought to involve strains of Escherichia coli. We report here that two wild-type Afa/Dr diffusely adhering E. coli (DAEC) strains, C1845 and IH11128, which harbor the fimbrial F1845 adhesin and the Dr hemagglutinin, respectively, and the E. coli laboratory strain HB101, transformed with the pSSS1 plasmid to produce Afa/Dr F1845 adhesin, all induced interleukin-8 (IL-8) production and transepithelial migration of polymorphonuclear leukocytes (PMNL) in polarized monolayers of the human intestinal cell line T84 grown on semipermeable filters. We observed that after PMNL migration, expression of decay-accelerating factor (DAF, or CD55), the brush border-associated receptor for Afa/Dr adhesins, was strongly enhanced, increasing the adhesion of Afa/Dr DAEC bacteria. When examining the mechanism by which DAF expression was enhanced, we observed that the PMNL transepithelial migration induced epithelial synthesis of tumor necrosis factor alpha and IL-1␤, which in turn promoted the upregulation of DAF.

Here, we provide evidence that disruption of the microtubule structure rapidly triggers extracellular signal-regulated kinase (ERK) activation, whereas it was without effect on SAPK2 activity, which is commonly acknowledged to control pro-inflammatory cytokine production. This process involves the activation of the entire cascade including Ras, Raf-1, MEK1/2, ERK1, and ERK2. Activation of ERKs is followed by their nuclear translocation. Although other SAPK congeners might be activated upon microtubule depolymerization, the activation of ERK1 and ERK2 is mandatory for IL-1 production as shown by the blocking effect of PD 98059, a specific MEK1/2 inhibitor. Additionally, we provide evidence that microtubule disruption also induces the activation of c-Src and Hck activities. The importance of Src kinases in the mediation of the colchicine effect is underscored by the fact that CP 118556, a specific inhibitor of Src-like kinase, abrogates both the colchicine-induced ERK activation and IL-1 production. This is the first evidence that ERK activation is an absolute prerequisite for induction of this cytokine. Altogether, our data lend support to a model where the status of microtubule integrity controls the level of Src activities that subsequently activate the ERK kinase cascade, thus leading to IL-1 production.

SUMMARYThe frequency of mononuclear cells (MNC) spontaneously secreting interferon-gamma (IFN-y) has been examined in freshly isolated cell suspensions from human palatine tonsils. Two-site reverse enzyme-linked immunospol (ELISPOT) analyses, involving short term (20 h) incubation of MNC in the absence of any added exogenous stimulus, revealed that tonsillar MNC suspensions contain exceptionally large numbers ofcells secreting IFN-y. No significant differences were observed when comparing the frequency of IFN-y-producing cells between cell suspensions obtained from hyperplastic and tonsillitis specimens. Cell-sorting experiments disclosed that spontaneous tonsiltar IFN-y production was essentially contributed by CD4"' T cells, and required the presence of accessory cells and/or soluble factors to be detected. Thus, depletion of plastic adherent cells or monocytes from the tonsillar MNC suspensions resulted in reduced numbers of detectable IFN-ysecreting cells. Addition of very small numbers of autologous monoey tes restored spontaneous I FN-y production in tonsillar MNC cultures depleted of monocytes. Neutralization of endogenous IL-l/J and IL-2. as well as blocking of the IL-2 receptor, also decreased IFN-y production from un fractionated tonsillar cells. Addition of exogenous IL-l/i restored IFN--; production in cultures of tonsillar MNC depleted of plastic adherent cells. Furthermore. IL-l/f synergized with lL-2 by increasing intracellular as well as cell-free levels of IFN-y in cultures of unfraclionatcd tonsillar MNC. This study further establishes that the tonsils are highly active immunologieal organs containing large numbers of T cells spontaneously producing lFN-y whose detection is contingent upon the presence of functional accessory cells. It also demonstrates that concomitant production of IL-1/? and IL-2 occurs in tonsils and is necessary to maintain ongoing synthesis and extracellular aecumulation of IFN-y in these organs.
