Detection of homology to the beta bacteriophage integration site in a wide variety of Corynebacterium spp

Abstract: In toxigenic conversion of Corynebacterium diphtheriae C7, ,B bacteriophage DNA integrates into either of two chromosomal attachment sites, aitBi or attB2. These attB sites share a 96-base-pair sequence with the attP sites of "-related phages. The distribution of attB-related sites in other species of Corynebacterium was assessed by hybridization with a DNA probe containing both attB sites of the C7 strain and a second DNA probe containing the attP site of a 13-related phage. All but one of the 15 C. diphtheri… Show more

“…Moreover, some strains of C. ulcerans and C. pseudotuberculosis can produce diphtheria toxin (DT). Furthermore, some non-toxigenic strains are converted to toxigeny (DT production) by β-phages from toxinogenic C. diphtheriae [15,23,24,44].…”

“…More specifically, each gene encoded a recombinase protein comprising 408 amino acids, with 11 (highly conservative) amino acid differences over their total lengths. The attB and attP regions from NCTC13129 and β are identical over the first 50 bp and in total share a 92 bp common 'core' region (Cianciotto et al, 1986): in addition, the core regions of β attP and the NCTC13129 prophage attP (deduced from attL1 and attR1) were identical over this region.…”

“…The β attB site of C. diphtheriae is present in other Coryneform species (Cianciotto et al, 1986), opening up the possibility that the vectors developed here could be used to transform other species of Corynebacterium. To test this hypothesis we focused on two species: C. glutamicum and C. ulcerans.…”

“…These attB sites share a 96-bp sequence with the attP sites of ␤-related phages (37). Moreover, screening of a variety of Corynebacterium species including the soil isolate C. glutamicum ATCC 13032 with attP and attB probes showed that all of the species investigated contained at least one DNA fragment that hybridized with both of these probes (8,9,15). Besides this report, information on the molecular biology of lysogeny in coryneform bacteria remained scarce.…”
