Detection of genetic divergence among some wheat (Triticum aestivum L.) genotypes using molecular and biochemical indicators under salinity stress

Abstract: Wheat has remarkable importance among cereals in Egypt. Salt stress affects plant growth, development, and crop productivity. Therefore, salinity tolerance is an essential trait that must be incorporated in crops. This research aimed to investigate molecular and biochemical indicators and defence responses in seedlings of 14 Egyptian wheat genotypes to distinguish the most contrasting salt-responsive genotypes. Analysis of ISSR and SCoT markers revealed high polymorphism and reproducible fingerprinting profile… Show more

“…It is clear that the total carbohydrate content was higher at 50 mM NaCl (331.309 mg/gDW). Whereas total carbohydrate content dropped at 150 and 200 mM NaCl by 262.65 and 224.68 mg/gDW, respectively, which is agreed with Gowayed and Abd El-Moneim (2021) results, who stated that total carbohydrate content has not changed at 150 mM NaCl. Moreover, Ali et al (2021) recorded a reduction in the total carbohydrate in the saline condition, which agrees with our results.…”

“…Remarkably, Sohag-5 was characterized as salt susceptible. The same genotype was previously reported as a salt susceptible genotype at the germination stage by Gowayed and Abd El-Moneim ( 2021 ).…”

“…Abou-Deif, et al, [54] used the ISSR molecular marker to estimate the genetic diversity between some Egyptian wheat genotypes. But El-Moneim, [55] and Gowayed and El-Moneim [9] used ISSR and SCoT primers to determine and evaluate some Egyptian wheat genotypes tolerant to drought and salt stress, respectively. But our current study used different primers for SCoT in genetic diversity and evaluation of Egyptian wheat genotypes' resistance to stem and leaf rust diseases.…”

“…Molecular marker techniques are considered a useful tool for the determination, characterization, and assessment of genetic diversity among different organisms at the species-level (Hanaa, et al 2020). SCoT and SRAP markers are effectively used in various species of microorganisms to test the genetic diversity, structure, identification of cultivars, quantitative trait loci (QTL) mapping and DNA fingerprinting [8,9]. However, the genetic diversity assessment between and within the species of Carthamus was carried out by SRAP [10,11].…”
