Detection of Dye-linkedD-Mannitol Dehydrogenase Activity inAcetobacter xylinumKU-1

Abstract: We detected dye-linked D-mannitol dehydrogenase activity in the crude extract of Acetobacter xylinurn KU-l. The enzyme activity was specific for D-mannitol, and not pyridine nucleotide (NAD + , NADP+)-dependent. The optimal pH was found to be 5.0, while the optimal temperature was at. 50°C. The enzyme activity was inhibited by p-quinone noncompetitively.

“…xylinum KU-I was obtained from the type culture collection of the Laboratory of Applied Biological Chemistry, Yamaguchi University, Japan. 4 ) A. xylinum KU-I was grown on the standard medium containing 2.0% (w/v) D-mannitol, 0.5% yeast extract (Wako Pure Chemical Industries Ltd., Osaka, Japan), 0.5% Polypeptone (Wako Pure Chemical Industries Ltd., Osaka, Japan), 0.27% disodium phosphate anhydrous, and 0.115% citric acid monohydrate in deionized water (pH 6.0). D-Mannitol was sterilized separately with a 0.2 J1.m filter unit (Kurabo Co., Tokyo, Japan), and added to the medium.…”
