Detection of Cyprinid Herpesvirus-3 in Field Samples of Common and Koi Carp by Various Single-Round and Nested PCR Methods

Abstract: Cyprinid herpesvirus-3 (CyHV-3, previously described as koi herpesvirus) is a causative agent of the lethal disease in koi and common carp that can adversely affect common and koi carp breeds. Outbreaks with mass mortality have occurred world wide since 1998. At present, several different polymerase chain reaction (PCR) tests are in use to detect CyHV-3 in koi and common carp. In this study, we compared the sensitivity of the single-round PCR assays of Gray, Gilad, and Bercovier, and further the Gilad and Berc… Show more

“…In Europe, reports of widespread mass mortality have been notified in carp farms and fisheries in Germany, Poland, and the UK (Bergmann, Kempter, Sadowski, & Fichtner, 2006;Gotesman, Kattlun, Bergmann, & El-Matbouli, 2013;Taylor, Dixon, et al, 2010). The disease is also known to occur in, or has been recorded in fish imported into, Austria, Belgium, Czech Republic, Denmark, France, Hungary, Italy, Luxembourg, The Netherlands, Republic of Ireland, and Switzerland (Haenen et al, 2004;McCleary et al, 2011;Pokorova et al, 2010;Pretto et al, 2013). Most recently, KHVD outbreaks have been reported to the OIE from Romania, Slovenia, Spain, and Sweden (OIE, 2012).…”

“…The internal primer set was used according to D. Stone (described in the CyHV-3 protocol distributed by CEFAS) and result in a final amplicon of 348 bp. The sequences of this primer set were according to Pokorova et al (2010). For preparation of the PCR reaction mixture, PPP master mix (TopBio, Czech Republic) was used.…”

“…Reaction mixtures (25 µl for both conventional and nested PCRs) consisted of 12.5 µl of PPP master mix, 9.5 µl of PCR water (TopBio, Czech Republic), 1 µl of DNA template, 1 µl of forward primer (0.1 µM) and 1 µl of reverse primer (0.1 µM). Cycling conditions for conventional and nested PCRs were according to Pokorova et al (2010). The nested PCR described above was used only in the no-stress experiment.…”

“…The nested PCR uses primers based on the sequence of the thymidine kinase gene of CyHV-3 (Bercovier et al 2005) supplemented with a nested primer set provided by D. Stone (Pokorova et al 2010) which is still recommended for routine diagnostics. The first set of outer primers was used according to Bercovier et al (2005) and give a final amplicon of 409 bp.…”
