Background and Aim:Animal chlamydiosis, caused by different chlamydial species, is characterized by clinical or subclinical disease manifestations in cattle, buffalo, ovine, caprine and wild animal species. Animal chlamydiosis often remains underdiagnosed or undiagnosed, and its status in many parts of India is still unknown. Hence, the present study was conducted to determine the seroprevalence of animal chlamydiosis amongst ruminant livestock species of five states of India.Materials and Methods:Totally, 2127 randomly selected serum samples collected from ruminant livestock species viz. cattle (n=430), buffaloes (n=429), sheep (906) and goats (n=362), were tested by agar gel precipitation test for chlamydiosis between 2002 and 2011. Precipitating antigen was prepared from locally isolated strain of Chlamydia psittaci after treatment with sodium deoxycholate.Results:The chlamydial seroprevalence detected amongst ruminants in five states of India was: Himachal Pradesh: Cattle-10.90%, sheep-10.60% and goats- 22.46%; Punjab: Cattle-1.45%; Andhra Pradesh: Cattle-2.80%, buffaloes-0.93%, sheep-8.90% and goats-9.46%; Maharashtra: goats-8.33%; Jammu and Kashmir: sheep-12.50%. The mean seroprevalence values of each animal species are: Cattle-4.65%, buffaloes-0.93%, sheep-9.82% and goats-19.33%.Conclusion:The results indicate the endemic nature of animal chlamydiosis across five states in India. Hence, it requires further extensive studies in other parts of India also using chlamydial species-specific diagnostics to ascertain overall countrywide prevalence of the disease. The zoonotic nature of the chlamydiae of ruminant origin further adds significance to such prevalence studies.

Summary Enzyme Immunoassay for Detection of IgG‐ and IgM‐Antibodies against Chlamydia psittaci in Pigeon Sera The development of an indirect ELISA for chlamydial antibodies in pigeon sera, including the production of the necessary immunoreagents, is described in the test procedure. The (H + L)‐chain specific anti‐IgG‐horseradish‐peroxidase conjugate produced could be diluted 1/10,000, and the H‐chain specific anti‐IgM conjugate 1/800. In comparative investigations with 219 pigeon sera, the ELISA was highly superior to the complement fixation (CF) reaction with regard to sensitivity, specificity, practicability, and reliability. Apparently, chlamydia‐specific IgM did not fix guinea pig complement in amounts detectable in the CF test. In contrast, the (H + L)‐chain specific ELISA system detected all immunoglobulins. This hypothesis was confirmed by simultaneous application of the CF test, the IgG‐ and IgM‐ELISA to a further 100 sera, as well as sera from experimentally infected pigeons. The possibility for a diagnosis of inapparent persistent chlamydial infection by detection of persisting IgM antibodies is discussed. Résumé Immuntest enzymatique por la mise en évidence des anticorps IgG et IgM contre Chlamydia psittaci chez le pigeon On décrit le développement d'un micro‐enzym‐immunassay (ELISA) pour la mise en évidence d'anticorps contre des chlamydias dans des sérums de pigeons ainsi que les réactifs immunologiques nécessaires à cet examen. Le conjugué H + L‐ chaînes anti‐IgG spécifique‐peroxidase du raifort a été dilué 1/10,000 et le conjugué H‐chaînes anti‐IgM‐conjugué a été dilué 1/800. Des recherches comparées sur 219 sérums de pigeons ont montré que l'ELISA surpassait la réaction de fixation du complément (CF) en sensibilité, spécificité, technique et fiabilité. Il est apparu que les immunglobuines de type IgM spécifiques n'étaient pas liées par le complément de cobaye. Le système ELISA‐chaînes spécifiques H + L a par contre permis la mise en évidence de toutes les immunglobulines. Cette hypothèse a été confirmée par l'emploi simultané de CF et ELISA anti‐IgG et anti‐IgM avec 100 autres sérums et avec des sérums recueillis dans un essai d'infection. On discute la possibilité de diagnostiquer des infections chroniques et inapparentes à Chlamydias avec ELISA par la mise en évidence d'IgM spécifiques et persistantes.
