Detection of benzo[a]pyrene diol epoxide-DNA adducts in embryos from smoking couples: evidence for transmission by spermatozoa

Abstract: Tobacco smoking is deleterious to reproduction. Benzo[a]pyrene (B[a]P) is a potent carcinogen in cigarette smoke. Its reactive metabolite induces DNA-adducts, which can cause mutations. We investigated whether B[a]P diol epoxide (BPDE) DNA adducts are detectable in preimplantation embryos in relation to parental smoking. A total of 17 couples were classified by their smoking habits: (i) both partners smoke; (ii) wife non-smoker, husband smokes; and (iii) both partners were non-smokers. Their 27 embryos were ex… Show more

“…DNA adduct levels at these time points did not differ between Xpc 2/2 and Wt mice, which is logical because they both lack the ability to remove DNA damage in the period between exposure and collection. Moreover, these spermatozoa did not divide, and it is important to note that at this stage DNA damage is present until after fertilization [Zenzes et al, 1999b]. Mutations may be induced after DNA replication resumes which, of course, holds for all DNA adducts present in any mature spermatozoa.…”

“…in fathers who smoke, can be passed on to their children and has been linked with an increased incidence of childhood cancer in their offspring (Zenzes et al 1999, Zenzes 2000. This has caused concern over the use of sperm from subfertile men in assisted reproductive technologies (IVF and ICSI).…”

“…DNA damage rates in sperm are higher in infertile individuals (Irvine et al 2000), patients subjected to cancer therapies (Charak et al 1990, Kenney et al 2001, men who smoke (Zenzes et al 1999, Zenzes 2000 and also possibly in older men (Wyrobek et al 2006). Concerns have been expressed that using sperm with damaged DNA for intracytoplasmic sperm injection (ICSI) could have a long-term effect on the health of the children born to subfertile couples (Gosden et al 2003).…”
