“…As found by Nassau et al (1975) and confirmed here, a serious difficulty with the indirect technique is adsorption of non-antibody globulin by antigen or supporting medium, or both, and this subsequently combines with labelled antiglobulin. In our assay system, the response of adsorbed globulin was equivalento 5-10 pg of antibody per ml serum.…”

“…Of the several different principles that can be used as a basis for immunoradiometric assay, two were selected for rigorous assessment. These were, first, competition between antibody in the test sample and added isotopically-labelled homologous antibody for attachment to antigen and, second, attachment of antibody in the test sample to solid-phase antigen, followed by reaction of the resulting complex with isotopically-labelled homologous anti-globulin (Rosenthal, Hayashi and Notkins, 1972;Nielsen, Parratt and White, 1973;Restall, Thomas and Pennant, 1973;Smith, Gehle and McCracken, 1974;Nassau, Parsons and Johnson, 1975). When assay procedures based on the two principles were applied to samples containing known concentrations of immunopurified antibacterial globulins in saline-phosphate buffer, sensitive detection and reproducible results were obtained.…”

“…Many authors have reported serological tests which have appeared reliable when employed against sera from patients with known diagnoses (Middlebrook and Dubos, 1948;Parlett and Youmans, 1959;Takahashi, 1962;Lind, 1964;Duboczy and White, 1966;Wallace et al, 1966;Cole, Lazarus, and Hedrick, 1972; Affronti, Fife, and Grow, 1973;Nassau, Parsons, and Johnson, 1975;Nicholls, 1975) but none of these has proved of sufficient clinical usefulness to become universally adopted.…”

“…Studies on the serology of tuberculosis began with the applications of the complement fixation test in 1901 [2], and extensive studies were introduced such as hemagglutination test [3j, precip itation and gel diffusion [4], the fluorescent antibodies test [5], radioimmunoassay [6], and enzyme-linked immuno sorbent assay (ELISA) [7]. Dicna [8] summarized the re quirements for the ideal serological test for tuberculosis.…”
