Detection of Anti-Sp100 Antibodies in Primary Biliary Cirrhosis. Comparison of Elisa and Immunofluorescence

Abstract: Primary biliary cirrhosis (PBC) is, which a chronic, autoimmune liver disease. Some patients have antinuclear antibodies anti-Sp100, which are considered to be disease-specific. We compared an enzyme-linked immunosorbent assay (ELISA) and indirect immunofluorescence (IIF) for detection of anti-Sp100. The sensitivity of anti-Sp100 determined by ELISA and IIF was 44% and 34%, respectively. Specificity was 99% for ELISA and 98% for IIF, respectively. The positive and negative predictive value (PPV, NPV) for anti-… Show more

“…They are moderately specific for PBC and can rarely be detected in rheumatological diseases and in acute hepatitis of any origin ( 33 , 98 , 99 ). Anti-sp100 result in a typical pattern of “nuclear dots” in IFT on HEp-2-cells ( 100 ); a confirmatory test (ELISA and Western Blot) is available ( 101 ).…”

“…The diagnostic and clinical relevance of anti-Sp100 antibodies in PBC has been extensively studied in recent years, but a correlation between levels of anti-PML or anti-Sp140 and PBC was studied much less frequently and has still not been fully defined [ 8 , 17 , 21 , 22 , 32 , 33 ].…”

“…Elevated serum concentrations of ANAs should be found in approximately 50% of PBC patients and 85% of AMA-negative PBC patients[96]. In short, 44% of PBC patients had anti-sp100, 15.1% had PML, 25% had anti-gp210 and 25% had ACAs[97-100]. AMAs and ANAs (anti-gp210, anti-sp100, ACAs) are particularly prevalent in PBC[101].…”
