Detection of an oestrogen receptor-like protein in human meningiomas by band shift assay using a synthetic oestrogen responsive element (ERE)

Abstract: When a ligand binding or enzyme immunoassay is used, meningiomas are found to be rich in progestin receptors (PR) whereas oestrogen receptors (ER) are virtually undetectable. A protein that can bind to an oestrogen responsive element (ERE) was detected in meningiomas, by using the band-shift assay. The binding of ER to the ERE is inhibited by the anti-ER monoclonal antibody ER-P31, which is directed to the A/B domain of the ER, indicating that the binding protein is an ER-like protein. “…The morbidity and mortality of benign forms of meningiomas can be influenced by other factors, mostly the tumour location and the radicality of resection, age and comorbidities [ 8 , 15 , 16 ]. Some authors have proven specific female-associated factors that increase the incidence of meningiomas in female population, which we confirmed also in our cohort [ 17 , 18 ]. Also, several genetic changes could play an important role in meningioma pathogenesis [ 19 ].…”

“…22 Another indirect argument for a biological role of variant ER transcripts is the correlation between their demonstration in tissues that are ER negative by binding assay and the presence of products of estrogen-responsive genes such as progesterone receptors 16,23 or pS2. 24 The presence of proteins capable of binding specifically to estrogen-responsive elements was shown by gel-mobility shift assay in certain ER-negative breast cancers 25 and in meningiomas 26 ; in the latter, the protein, different from the wild type ER, was recognized by antibodies specific for a region different from the hormone-binding domain of the receptor. A surprising correspondence with the events we have shown in humans has been shown in rats treated with peroxisome proliferators 27 : in this experimental model, which closely resembles human carcinogenesis, there FIG.…”
