Detection and identification of furan fatty acids from fish lipids by high-performance liquid chromatography coupled to electrospray ionization quadrupole time-of-flight mass spectrometry

Abstract: Using high-performance liquid chromatography coupled to high-resolution electrospray ionization quadrupole time-of-flight mass spectrometry (HPLC/ESI-Q-TOF-MS), we have developed a new method for detection and identification of furan fatty acids (F-acids), which are widely distributed in living organisms and foods as minor lipid components and are known to have antioxidant and anti-inflammatory effects. For this purpose, total fatty acids prepared from the testis lipids of Japanese chum salmon (Oncorhynchus ke… Show more

“…Namely, the first and the second number denote the number of carbon atoms of the carboxyalkyl and the alkyl chain, respectively, while the central letter indicates the methylation degree on the furan moiety ("M" = monomethyl-substituted in β-position, "D" = dimethyl-substituted in β,β′-positions) [4] (Table 1). For better readability in TAGs, numbers in these short forms (e.g., 9D5) will be presented in superscript and subscript style (e.g., 9 D 5 ) [6].…”

“…Development of various analytical technologies facilitates the identification and quantification of unknown and trace substances in complex samples. , Quadrupole-time-of-flight mass spectrometer, taken as an example, needs a small amount of sample for analysis with its well-known selectivity and sensitivity. Besides, this machine has developed several scan functions, as it shared a quadrupole, T-wave element and time-of-flight cells. The derived developed data-independent acquisitions, such as data-dependent acquisition (DDA) and data-independent mass spectrometry (MS E ), can collect all mass data of precursor and fragment ions fully. , In general, several acquisition modes provide a powerful platform for compound detection with rich chemical mass information.…”

“…Like m/z 123, m/z 279 was present in the GC/MS spectra of all three D-FuFA-MEs (Figure B–D) as the fragment ion m/z 279 is formed by a cleavage in the alkyl chain with only one carbon atom being left in the measured fragment. The presence of 9D4 in the fungi sample is remarkable because this FuFA had not been described before in soy, herbs and vegetables, butter or fish. ,, Hitherto, traces of 9D4 (0.35% of the total FuFA content) were only reported in hepatopancreas and in muscle phospholipids of one crustacean sample. In our study, 9D4 was detected in 11 of the 37 fungi samples, including seven Agaricus , two Pleurotus, and two meadow mushroom samples (Supporting Information, Table S2).…”
