Detection and fine‐mapping of SC7 resistance genes via linkage and association analysis in soybean

Abstract: Soybean mosaic virus (SMV) disease is one of the most serious and broadly distributed soybean (Glycine max (L.) Merr.) diseases. Here, we combine the advantages of association and linkage analysis to identify and fine-map the soybean genes associated with resistance to SMV strain SC7. A set of 191 soybean accessions from different geographic origins and 184 recombinant inbred lines (RILs) derived from Kefeng No.1 (resistant) × Nannong 1138-2 (susceptible) were used in this study. The SC7 resistance genes were … Show more

“…Unlike the Rsv1 and Rsv3 loci, the Rsv4 region lacked genes similar to known R genes, suggesting that Rsv4 functions via a potentially novel resistance mechanism (Ilut et al, 2016). Additional candidate SMV resistance genes on chromosome 2 have been identified for Rsv4 and for Rsc7, Rsc8 and Rsc18 (Klepadlo et al, 2017a;Li et al, 2015;Wang et al, 2011b;Yan et al, 2015;Zhao et al, 2016) (Table 1).…”

“…The Rsc-15 resistant gene was mapped between Sat_213 and Sat_286 with distances of 8.0 and 6.6 cM to the respective flanking markers on chromosome 6 (Yang and Gai, 2011). The resistance gene Rsc-7 in the soybean cultivar Kefeng No.1 was mapped to a 2.65 mega-base (Mb) region on soybean chromosome 2 (Fu et al, 2006) and was subsequently narrowed down to a 158 kilo-base (Kb) region (Yan et al, 2015). Within 15 candidate genes in the region, one NBS-LRR type gene (Glyma02g13600), one HSP40 gene (Glyma02g13520) and one serine carboxypeptidase-type gene (Glyma02g13620) could be the candidates for Rsc-7 .…”

“…Charcoal rot QTL on chromosome 15 is located downstream of the Fusarium graminearum resistance QTL mapped close to BARC-042629-08331 (Gm15:01276087) in soybean cultivar Conrad (Ellis et al, 2012), and Soybean mosaic virus strain SC7 resistance mapped close to BARC-018959-03045 (Gm15:01303462) in Kefeng No.1 (Yan et al, 2015). Also, the same QTL partially overlaps with the resistance QTL to Sclerotinia sclerotiorum, which causes Sclerotinia stem rot, mapped between Satt411 and Satt369 (Gm15:02517404-49011508) in PI 391589B (Guo et al, 2008).…”

“…The region on chromosome 16 between the two newly identified QTLs for charcoal rot include QTLs for resistance to Soybean mosaic virus strain SC7, mapped near BARC-041267-07957 (Gm16:31944251) in Kefeng No.1 (Yan et al, 2015), and to soybean cyst nematode cqSCN-003 (Heterodera glycines [HG] Ichinohe) race 3 (HG Type 7), mapped between Satt244 and Satt547 (Gm16:33819094-34035391) (Glover et al, 2004;Guo et al, 2005;Chang et al, 2011). Also, one QTL on chromosome 16 overlapped with soybean cyst nematode race 2 (HG Type 1.2.5.7.…”
