Detection and differential diagnosis of colon cancer by a cumulative analysis of promoter methylation

Abstract: Alterations in the methylation of promoters of cancer-related genes are promising biomarkers for the early detection of disease. Compared with single methylation alteration, assessing combined methylation alterations can provide higher association with specific cancer. Here we use cationic conjugated polymer-based fluorescence resonance energy transfer to quantitatively analyse DNA methylation levels of seven colon cancer-related genes in a Chinese population. Through a stepwise discriminant analysis and cumul… Show more

“…Previous work by the Wang research group used cationic conjugated polymers (CCP) as the energy donor to quantitatively detect gene methylation and achieve high sensitivity for colon cancer detection (Feng et al, 2008(Feng et al, , 2010Yang et al, 2012;Zhang et al, 2014). When compared with organic fluorophores, QDs have several advantages, including a narrow emission spectrum, broad absorption spectrum, negligible photobleaching, and high photochemical stability Clapp et al, 2006;ReschGenger et al, 2008;Hötzer et al, 2012;Su et al, 2014).…”

“…222, 225, 226 Indeed, nano-cytology and nano-cytoarchitecture have been proposed as screening targets for field carcinogenesis. 226, 227 It is therefore conceivable that the sophistication of future biosensors will enable accurate assessment of in-vivo real-time changes in the microenvironmental interactome with high resolution (i.e., in-vivo molecular pathology).…”

“…Methylated DNA is another promising biomarker in urine or blood that can be used for early detection of cancer and other genetic diseases [276,277]. Quantitation of cellular DNA methylation levels in colon cancer patients has shown to be a promising alternative to colonoscopy for detecting colorectal cancer [278].…”
