Detection and Control of Fusarium oxysporum from Soft Rot in Dendrobium officinale by Loop-Mediated Isothermal Amplification Assays

Abstract: Soft rot causing Fusarium oxysporum is one of the most destructive diseases of Dendrobium officinale Kimura et Migo in China that reduces D. officinale yield and quality. A key challenge for an integrated management strategy for this disease is the rapid and accurate detection of F. oxysporum on D. officinale. Therefore, a new loop-mediated isothermal amplification (LAMP) assay was developed for this purpose. In this study, the primers were selected and designed using the translation elongation factor-1α (TEF-… Show more

“…Based on this, the LAMP technology is a good strategy for the early diagnosis of bacterial diseases, which can prevent plant disease and pathogens during the preliminary stage and aids in health control and yield optimization without depending on the pest and pesticides usage (Le and Vu, 2017 ). Xiao and Li ( 2021 ) detected the soft rot pathogen of Dendrobium officinale by the LAMP method and then tested the control effect of several fungicides. After 14 days of spraying, the synergistic efficacy of the two fungicides (pyraclostrobin and picoxystrobin) reached 82.39%.…”

“…With the development of D. officinale industrialization, it is urgent to improve the resistance of tissue culture seedlings by genetic engineering. It is common for D. officinale to be exposed to various pathogens, including fungus, bacteria, and viruses during industrial cultivation [6][7][8][9]. For example, the common and destructive fungal pathogen, F. oxysporum, always causes stem rot of D. officinale and has a high incidence of 30% to 50% [8,40].…”

“…When compared with conventional PCR, LAMP offers obvious advantages of visualisation through turbidity or colour changes [ 27 , 34 , 35 ]. LAMP technology is currently widely used for the rapid detection of pathogens such as fungi [ 36 , 37 , 38 , 39 ], bacteria [ 40 , 41 , 42 ] and viruses [ 43 , 44 ].…”
