Detailed investigation of the diagnostic value in tumour histopathology of ICR.2, a new monoclonal antibody to epithelial membrane antigen

Abstract: The production and detailed immunostaining properties of a new rat monoclonal antibody (ICR.2) to epithelial membrane antigen are reported. The antibody was selected for its ability to compete with the polyclonal antiserum (M7), used in the original immunohistological studies, in order that it might serve as a direct replacement in diagnosing epithelial tumours. Most of the staining reactions on normal tissues were identical to those previously reported with M7 but there were some important differences. They i… Show more

“…Antibodies to the epithelial membrane antigen (EMA) [20,21,381 have been used to label luminal cells, and the common acute lymphoblastic leukemia antigen (CALLA/CDl O/neutral endopeptidase 24.1 1) was used to mark the myoepithelial cells [17]. We report here the appearaucc.…”

“…The total absence of skin involvement and the partial clear cell pattern speaks against spindle-cell melanoma [3]. Besides the identification by the protein S100 [1,3] other new techniques are now used to confirm the diagnosis of CCS such as ICR2 [5], a new monoclonal antibody to epithelial membrane antigen and HMB-45 [4] a melaninassociated antigen. On the other hand, cytogenetic techniques [2] are reported to demonstrate clonal, numerical and structural abnormalities with specific involvement of chromosome 2 and 22 in CCS.…”

“…The presence of lymph node metastasis and intracellular glycogen speaks in favor of malignant melanoma. However, the presence of melanocytes in tendons and aponeurosis has never been recognized [5]. The positive reaction with PS 100 is additional proof in favor of malignant melanoma and clear cell sarcoma.…”
