Design, Synthesis, and Evaluation of Dihydrobenzo[cd]indole-6-sulfonamide as TNF-α Inhibitors

Abstract: Tumor necrosis factor-α (TNF-α) plays a pivotal role in inflammatory response. Dysregulation of TNF can lead to a variety of disastrous pathological effects, including auto-inflammatory diseases. Antibodies that directly targeting TNF-α have been proven effective in suppressing symptoms of these disorders. Compared to protein drugs, small molecule drugs are normally orally available and less expensive. Till now, peptide and small molecule TNF-α inhibitors are still in the early stage of development, and much m… Show more

“…Numerous functions have been described for TNF, a ubiquitously expressed proinflammatory cytokine ( Kroetsch et al, 2017 ) supporting the inflammatory response ( Deng et al, 2018 ). Increased expression of TNF is associated with osteogenesis of the LF, which contributes to spinal stenosis ( Lin et al, 2018 ).…”

“…Research suggests that in order to inactivate TNF-α trimer, the complex may form an intermediate form that can undergo subunit dissociation. Herein, we have targeted TNF-α dimer to prevent its trimeric association, thereby inhibiting its function [63][64][65][66].…”

“…However, it will not only stimulate glial cells to excessively secrete tumor necrosis factor 2 BioMed Research International TNF-α but also promote peripheral inflammatory cytokines transported from the blood into the brain when the integrity of the blood-brain barrier is impaired. The high level of tumor necrosis factor associated with the inflammatory response after stroke is an important cause of poststroke epilepsy [37]. Studies have noted increased TNF-α serum levels in individuals with epilepsy compared with normal subjects.…”
