Design, Synthesis, and Cercaricidal Activity of Novel High‐efficient, Low‐toxic Self‐spreading PEGN‐salicylanilide Derivatives Against Cercariae Larvae of Schistosome Japonicum Floating on the Water Surface

Abstract: Novel cercaricides of PEG-N-salicylanilide derivatives that could self-spread and float on the water surface were designed and synthesized according to the particular habit of cercariae larvae of Schistosome japonicum. The structures of the cercaricides were characterized by the infrared spectra (IR), magnetic resonance ((1) H NMR), and mass spectrum (MS). The images of the floating cercaricides on the water surface were investigated by the Brewster angle microscopy (BAM). When the cercaricides were dropped on… Show more

“…Six species of the genus Schistosoma including S. japonicum , S. mansoni , S. haematobium , S. mekongi , S. malayensis , and S. intercalatum can cause disease in people (Gryseels et al 2006 ). The life cycle of the parasite is complex with an intermediate host ( Oncomelania snails), definitive host (humans and mammals), and seven lifecycle stages involving the adult worm, egg, miracidium, mother sporocyst, daughter sporocyst, cercariae, and schistosomulum (Colley et al 2014 ; Guo et al 2008 , 2015 ). It is a great challenge to control schistosomiasis in undeveloped regions because of financial burden and environmental pressures.…”

“…It exhibited strong to moderate cytotoxicity on several human cell lines (12,13). In the last 2 decades, most of the modifications of niclosamide have focused on improving its solubility by altering formulations (e.g., as wettable powder, suspension concentrate, and polymeric controlled-release formulations) (14)(15)(16)(17) or by linking it with surfactants (e.g., polyethylene glycols [PEGs]) (18)(19)(20)(21). To the best of our knowledge, little effort has been made to reduce its toxicity through structure modification.…”

“…Schistosoma cercariae are (a non-feeding infective stage) penetrate and reproduce within a host of the class Mammalia [20]. Among the species of Schistosoma which may infect humans, the most harmful are Schistosoma mansoni , Schistosoma japonicum and Schistosoma haematobium , which together comprise over 95% of human infections [25].…”
