Design of novel multifunctional targeting nano-carrier drug delivery system based on CD44 receptor and tumor microenvironment pH condition

Abstract: In this study, to develop a multifunctional targeting nano-carrier drug delivery system for cancer therapy, the novel pH-sensitive ketal based oligosaccharides of hyaluronan (oHA) conjugates were synthesized by chemical conjugation of hydrophobic menthone 1,2-glycerol ketal (MGK) to the backbone of oHA with the histidine as the linker of proton sponge effect. The multifunctional oHA conjugates, oHA-histidine-MGK (oHM) carried the pH-sensitive MGK as hydrophobic moieties and oHA as the target of CD44 receptor. … Show more

“…Many research groups investigated the ability of nanoparticles (Salem, 2010;Abdelbary et al, 2015) and nanovesicles (Abdelbary & El-Gendy, 2008;Abraham lingan et al, 2011) to deliver drugs more effectively into targeted sites. Nanoparticles' ability to treat tumors has been extensively investigated by different scholars (Paciotti et al, 2004;Chen et al, 2016) Solid lipid nanoparticles are colloidal particles, consisting lipid materials, which are stabilized with the use of varying types of surfactants and cosurfactants. Their use is mainly in topical route (Ü ner et al, 2014;Samein 2014).…”

“…/10717544.2016 influenced by the tumor microenvironment, which is characterized by hypoxia and low pH (acidic cellular environment). Acidic pH outside cells limits uptake of weak base drugs such as doxorubicin, adriamycin and mitoxantrone (Chen et al, 2016). HIF-1a is a hypoxia-activated transcription factor that upregulates target genes by binding to the Hypoxia Response Element (HRE).…”

“…It is the third most common cancer and the fourth leading cause of mortality among cancer patients. Nanodrug delivery systems have been widely recognized as an emerging and promising field with the potential to offer solutions to the current obstacles in cancer therapies (Farokhzad & Langer, 2009;Zhao et al, 2013;Chen et al, 2014). Moreover, targeting nanocarriers could further improve nanocarriers selective delivery to the tumor tissue.…”
