Design of Hybrid Polymeric-Lipid Nanoparticles Using Curcumin as a Model: Preparation, Characterization, and In Vitro Evaluation of Demethoxycurcumin and Bisdemethoxycurcumin-Loaded Nanoparticles

Abstract: Polymeric lipid hybrid nanoparticles (PLHNs) are the new generation of drug delivery systems that has emerged as a combination of a polymeric core and lipid shell. We designed and optimized a simple method for the preparation of Pluronic F-127-based PLHNs able to load separately demethoxycurcumin (DMC) and bisdemethoycurcumin (BDM). CUR was used as a model compound due to its greater availability from turmeric and its structure similarity with DMC and BDM. The developed method produced DMC and BDM-loaded PLHNs… Show more

“…3 Different superscript letters in the same column or different superscript signs in the same row indicate differences are significant (p < 0.05) using one-way analysis of variance (ANOVA) with a Tukey post hoc. 4 Results for DMC and BDM were previously reported by our group [50].…”

“…Comparing the spectra of CQ and LBN-CQ ( Figure 3 e), the disappearance of characteristic bands of CQ at 1576 cm −1 and 1041 cm −1 and band shifts from 1605 cm −1 to 1639 cm −1 and from 1200 cm −1 to 1217 cm −1 were observed. The disappearance or shifts of characteristic bands from CQ can indicate interactions of the drug with the nanosystem, reiterating its presence in the nanocarrier [ 62 , 63 , 64 , 65 , 66 ]. These findings, along with the high encapsulation efficiency, could be an indication of drug encapsulation in the nanoformulation [ 62 , 63 , 64 , 65 , 66 ].…”

(2022) [33] Ismail-Alhasawi (2022) [34] Fawzy et al On the other hand, an additional approach to solving the problem of poor solubility of curcuminoid-type molecules has been the discovery of new formulations, including liposomes [39,40], nanoparticles [41][42][43][44][45], or the preparation of inclusion complexes with cyclodextrins [46,47]. Beta-cyclodextrin (BCD) is the first choice [48] for the administration of drugs toward their sites of action because of its low toxicity [49], certain hydrophilicity [50], and adequate cavity.…”
