Design of a Compact Disk-like Microfluidic Platform for Enzyme-Linked Immunosorbent Assay

Abstract: This paper presents an integrated microfluidic device on a compact disk (CD) that performs an enzyme-linked immunosorbent assay (ELISA) for rat IgG from a hybridoma cell culture. Centrifugal and capillary forces were used to control the flow sequence of different solutions involved in the ELISA process. The microfluidic device was fabricated on a plastic CD. Each step of the ELISA process was carried out automatically by controlling the rotation speed of the CD. The work on analysis of rat IgG from hybridoma c… Show more

“…In the past, fluidic control included syringe pumps (11), hydrogel valves (12), gravity-driven pumps (13), evaporation-based pumps (14,15), acoustic pumps (16), gas-generationbased pumps (17,18), and centrifugal force in CD chips (19). Electrokinetic flow (20)(21)(22), thermopneumatic (23,24), pneumatic͞hydraulic (25), or mechanical (26,27) valves and pumps have a high degree of control, but require external connection lines to larger equipment for actuation.…”

“…For example, capillary burst valves prevent the circulation of the fluid forming a barrier when the channel section expands abruptly. Sandwich, direct and competitive immunoassays have been developed on CDs [153,154,155]. This makes these devices perfect candidates to be used in the field.…”
