Culicoides Latreille biting midges are important blood feeding insects. Many species are pests and vectors of the disease causing agents including viruses, protozoa and filarial nematodes which can be transmitted to humans and other animals. However, knowledge of the role of Culicoides as vectors of filarial nematodes is limited, particular in Thailand, where at least 100 species of the genus Culicoides have been reported. In this study, a molecular approach using the 12S rRNA gene sequence was used to detecting the filarial nematode in four common biting midge species, C. actoni Smith, C. oxystoma Kieffer, C. peregrinus Kieffer and C. mahasarakhamense Pramual, Jomkumsing, Piraonapicha & Jumpato in animal shelters from northeastern Thailand. A total of 1,721 specimens were used for molecular screening. An unidentified Onchocercidae sp. was detected in a specimen of C. mahasarakhamense collected from Maha Sarakham province. This filarial species shows 93% sequence similarity with an unidentified Onchocercidae sp. isolated from Culex mosquitoes. Phylogenetic analyses revealed that Onchocercidae sp. from C. mahasarakhamense formed a clade with strong bootstrap support (100%) with filarial species detected in birds. Thus, it is very likely that the Onchocercidae sp. found in this study employes birds as vertebrate hosts which agrees with feeding behavior of C. mahasarakhamense which is known to feed on chicken. Further study is requiring to examine whether this biting midge species is a competent vector of this Onchocercidae sp.

An adult Indian buffalo (Bubalus bubalis) presented with corneal opacity, irritation, and excessive lacrimation from the left eye in the Referral Veterinary Polyclinic-Teaching Veterinary Clinical Complex (RVC-TVCC), Indian Veterinary Research Institute, Izatnagar. Clinical examination revealed a whitish thread-like worm in the left eye’s anterior chamber. The worm was surgically removed from the eye with supportive nerve blocks. Light microscopy was used for parasite morphological identification, which provided insight into the worm as female Setaria sp. Genomic DNA was isolated, and polymerase chain reaction amplification of 12S rRNA was conducted for molecular confirmation of the parasite. The amplicon was sequenced and analysed by bioinformatics software. Sequence data showed an amplicon size of 243 bp. Phylogenetic analysis with reference data from the NCBI Genbank database revealed the worm was S. digitata, with a similarity of 99.17%. The common predilection site of S. digitata is in the peritoneal cavity of natural hosts like cattle and buffalo and is mostly non-pathogenic. The aberrant migration of the parasite larva to the brain and eye commonly occurs in goats, sheep, and horses, causing clinical conditions like cerebrospinal nematodiasis (lumbar paralysis) and ocular setariasis, respectively. Nevertheless, until now, there have been no reports of ocular setariasis in buffalo. This report is the first unusual occurrence of ocular setariasis in buffalo and its molecular confirmation and phylogenetic analysis using 12S rRNA.

Abstract. Hanafiah M, Athaillah F, Helmi TZ, Sutriana A. 2023. Morphology of Setaria spp. (Setariidae; Nematoda) in Aceh cattle, Indonesia. Biodiversitas 24: 4151-4160. Setaria spp. filaria usually inhabit the peritoneal cavity and its microfilaria can be found in cattle blood. Adult worms are considered non-pathogenic but may cause mild fibrinous peritonitis. However, microfilaria can cause serious conditions. The worms migrate randomly to the central nervous system in a non-natural host such as sheep, horses, and goats, causing serious neuropathological symptoms which are known as epizootic cerebrospinal setariosis or nematodiasis cerebrospinal. The objective of this research was to determine the prevalence of Setaria spp. microfilaria in the blood of infected Aceh cattle, identify the worm species in the peritoneal cavity, and determine the morphological difference of Setaria spp. through light microscope (LM) and Scanning Electron Microscope (SEM). This research was performed on 150 Aceh cattle slaughtered in Banda Aceh slaughterhouse. The hematocrit microcapillary method and modified Knott’s Technique were used to determine the presence of Setaria spp. microfilaria in blood, while LM and SEM were used to identify Setaria spp. adult worm. The results showed that Setaria spp. was detected in Aceh cattle blood based on modified Knott’s Technique and hematocrit microcapillary method with a similar prevalence of 13.3%. The worm identification revealed the presence of Setaria spp. in the peritoneal cavity of Aceh cattle. The morphological differences between Setaria digitata and Setaria labiatopapillosa were successfully discovered through LM and SEM studies.
