Description of Hymenobacter daejeonensis sp. nov., isolated from grass soil, based on multilocus sequence analysis of the 16S rRNA gene, gyrB and tuf genes

Abstract: A polyphasic taxonomic study was carried out on strains PB105 and PB108 isolated from a grass soil in Korea. The cells of the strains were Gram-stain negative, non-spore-forming, non-motile, and rod-shaped. Comparative 16S rRNA gene sequence studies showed a clear affiliation of these strains with Bacteroidetes, which showed high pairwise sequence similarities with Hymenobacter algoricola VUG-A23a (99.2%), Hymenobacter fastidiosus VUG-A124a (97.4%), and Hymenobacter daecheongensis Dae14 (96.9%). The phylogenet… Show more

“…The surface water samples were directly spread onto modified 1/10 Reasoner's 2A (R2A) medium [11] and incubated at 25 °C under heterotrophic conditions. One ivory colony, designated Gr-4 T , was isolated after 3 days using the dilution-to-extinction technique [12] and routinely subcultivated on R2A agar at 30 °C for 48 h and kept in a glycerol solution (20 %, v/v) at −70 °C for long-term preservation. For most experiments, all strains were cultivated on R2A agar (Difco) at 30 °C.…”

“…Strains con5 T and con4 were recovered from a culture of Anabaena using a serial dilution method [16]. For the Anabaena culture, Anabaena variabilis FBCC010004 strain was obtained from the Freshwater Bioresources Culture Collection (FBCC, https://fbcc.nibr.re.…”

“…Identification of the 16S rRNA gene sequence of strain Ji-3-8 T was performed using the EzBioCloud server [20]. The phylogenetic trees were reconstructed with mega version 7.0 as described by Kumar et al [21,22]. Whole genomes of strain Ji-3-8 and C. rhizosphaerae KCTC 52515 T were sequenced using both the SMRT (Single Molecule, Real-Time) platform at Novogene Biotechnology (Beijing, PR China) using Illumina next-generation sequencing technology.…”
