Description of an Ectothermic TCR Coreceptor, CD8α, in Rainbow Trout

Abstract: We have cloned the first CD8α gene from an ectothermic source using a degenerate primer for Ig superfamily V domains. Similar to homologues in higher vertebrates, the rainbow trout CD8α gene encodes a 204-aa mature protein composed of two extracellular domains including an Ig superfamily V domain and hinge region. Differing from mammalian CD8α V domains, lower vertebrate (trout and chicken) sequences do not contain the extra cysteine residue (C strand) involved in the abnormal intrachain disulfide bridging wit… Show more

“…However, recent evidence suggests that mechanisms of T cell activation and functionality in fish are similar to those observed in mammals based on the identification of T cell Ag receptor (16 -18), MHC (reviewed in Ref. 19), CD3 (19 -22), CD8␣ and CD8␤ (23,24), and LCK (25) in bony fish. Recently, a CD4-like molecule containing two Ig domains (IgV and IgC) was described in the sea lamprey Petromyzon marinus (26), and a four-domain CD4 molecule was identified in the pufferfish Takifugu rubripes (27).…”

“…Three types of full-length B7 cDNA were isolated from head kidney cDNAs by RACE with specific primers (supplemental Table I) 5 based on predicted B7 DNA sequences. Fugu B7 structures were analyzed with the following programs: SignalP (www.cbs.dtu.dk/services/SignalP) for signal peptide, SOSUI (sosui.proteome.bio.tuat.ac.jp/sosuimenu-0.html) for transmembrane region, and InterPro (www.ebi.ac.uk/interpro/) for Ig-like domains.…”

“…TCR-MHC-peptide interactions induce signaling events through CD3 in mammals and most likely in lower vertebrates because a complete set of CD3 genes, including TCR, CD3⑀, and CD3␥␦, has been found in Pufferfish and Xenopus (39,40). CD8␣ has been identified in rainbow trout (41); however, the cytoplasmic tail lacks the consensus p56 lck motif, suggesting that TCR/CD3/CD8-mediated signaling events may be different in teleosts.…”
