Derlin-1 is overexpressed in human breast carcinoma and protects cancer cells from endoplasmic reticulum stress-induced apoptosis

Abstract: Introduction Aberrant microenvironment and endoplasmic reticulum (ER) stress are associated with solid-tumor progression. Stress proteins, like heat shock proteins and glucose-regulated proteins, are frequently overexpressed in human tumors. It has been reported that derlin-1 is involved in ER stress response. In vitro studies have demonstrated that derlin-1 participates in the retrotranslocation of misfolded proteins from ER into the cytosol. Because the roles of derlin-1 in human cancer have not yet been cha… Show more

“…Derlin-1 was up-regulated by these treatments as well as GPR78 ( Supplementary Fig. S5), which is concordant with the result of Wang et al (9). Membrane location of Derlin-1 was frequently observed in the cancerous tissues but was seldom observed in nonmalignant tissues (Fig.…”

“…Derlin-1 expression is up-regulated by inducers of ER stress in yeast (8) and Caenorhabditis elegans (6) and suppresses ER stress-induced apoptosis in tumor cells (9). Almost all solid tumors encounter ER stress, such as hypoxia, and tumor growth depends on an intact unfolded protein response (10 -13).…”

“…NFB has a known role in PEL tumorigenesis. It is modulated by the KSHV homolog of the cellular FLICE inhibitory protein (vFLIP) 52,58 as well as KSHV K15. 59 Of note, the EBV latency membrane protein 1 (LMP-1) can also activate NFB.…”

“…DERL1 and ETV1 have previously been associated with viral and nonviral cancers. 51,52 DERL1 protects cells from endoplasmic reticulum (ER) stress-induced apoptosis and is overexpressed in breast cancer. 52 Multiple reports have shown associations between solid tumors and ETV1, and alteration in ETV1 is a prognostic marker in tumor progression.…”
